小柴胡汤不同煎煮方式对黄岑有效化学成分的影响

摘要 目的:探析不同煎煮方式對小柴胡汤中黄岑有效化学成分的影响。方法:选取高效液相色谱法测定小柴胡汤用去滓再煎法与不去滓煎法2种煎煮后的黄岑有效化学成分(黄岑苷、黄岑素、汉黄岑素)的含量。对照品溶液包括黄岑苷、黄岑素、汉黄岑素,检测开始前先进行对照品溶液的线性关系考察及精密度试验、稳定性试验、回收率试验。结果:黄岑苷、黄岑素、汉黄岑素对照品溶液的标准曲线方程分别为Y=4 345 730.98X-8 888.11(r=0.999 8)、Y=2 478 268.68X911.05(r=0.999 5)、Y=771 988 9.74X2 449.03(r=0.999 9)。黄岑苷、黄岑素、汉黄岑素对照品溶液的精密度试验、稳定性试验与回收率试验的RSD均低于2%,平均回收率均高于95%。供试品溶液A中所测得的黄岑苷、黄岑素、汉黄岑素含量分别为(0.364 2±0.027 1)g/L、(10.192 3±0.387 5)g/L、(18.082 2±0.746 5)μg/mL,高于供试品溶液B的(0.282 4±0.047 8)g/L、(8.215 0±0.354 1)g/L、(12.698 2±0.642 9)μg/mL(P<0.05)。结论:去滓再煎法较之传统不去滓煎法更利于小柴胡汤中黄岑有效化学成分黄岑苷、黄岑素、汉黄岑素的溶出。

Effects of Different Decoction Methods of Xiaochaihu Decoction on Active

Chemical Constituents of Scutellaria Baicalensis Georgi

Abstract Objective:To study the effects of different decoction methods on the effective chemical composition of Scutellaria baicalensis Georgi in Xiaochaihu Decoction. Methods:The contents of effective chemical composition (Baicalin, Baicalein, Wogonin) in Xiaochaihu decoction boiled by deslagging and non-deslagging and then decocting were determined by high performance liquid chromatography. The comparison solution included the Baicalin, Baicalein and Wogonin. The linear relationship of the comparison solution and the precision test, the stability test and the recovery rate test were carried out before the start of the test. Results:The standard curve equation of Baicalin, Baicalein and Wogonin comparison solution were Y=4 345 730.98X888 8.11 (r=0.999 8), Y=247 826 8.68X911.05 (r=0.999 5) and Y=771 988 9.74X244 9.03 (r=0.999 9). The RSD of Baicalin, Baicalein, Wogonin reference solution in the precision test, stability test and the recovery test were all lower than 2% and the average recoveries were all higher than 95%. The contents of Baicalin, Baicalein and Wogonin in test solution A were (0.364 2±0.027 1)mg/mL, (10.192 3±0.387 5)mg/mL and (18.082 2±0.746 5)μg/mL, which were higher than (0.282 4±0.047 8)mg/mL, (8.215 0±0.354 1)mg/mL and (12.698 2±0.642 9)μg/mL in test solution B (P<0.05). Conclusion:The deslagging and then decocting were more beneficial to the dissolution of Baicalin, Baicalein, Wogonin in Xiaochaihu decoction compared to the non-deslagging and then decocting.

小柴胡汤为临床经典方剂,在各种发热性疾病的治疗中应用广泛。“去滓再煎”是药物的一种特殊煎煮方式,即于药物煎煮一定时间后将药渣滤去再次煎煮使药液浓缩。有中医研究认为,去滓再煎可使药液浓缩,减少药液量,减轻患者胃肠负担;另外药汁再煎后药液浓度大,适用于水液运化、转输障碍的患者[1]。已有研究显示,去滓再煎法在半夏泻心汤、小柴胡汤等方剂中较为多用,且以上汤方均为临床常用方剂[2]。但有专家提出[3-4],煎煮方式、药物配伍比例、炮制等因素均会对中药方剂中有效成分的溶出造成影响,但有关去滓再煎、不去滓再煎后有效……
