辣椒素对变应性鼻炎大鼠鼻黏膜及P物质表达的影响研究

[摘要] 目的 探讨辣椒素对变应性鼻炎大鼠鼻黏膜形态、P物质表达的影响及作用机制。 方法 建立变应性鼻炎动物模型,采用辣椒素滴鼻,记录大鼠喷嚏次数、抓鼻次数、流涕情况评分叠加计分。对比正常组、未治疗组、治疗组鼻黏膜形态,对比治疗后鼻黏膜肥大细胞、P物质含量变化。 结果 治疗后治疗组变应性鼻炎症状明显缓解(P<0.05),治疗组肥大细胞、P物质含量较未治疗组明显减少(P<0.05)。 結论 辣椒素对变应性鼻炎有治疗作用,其作用机制与减少P物质含量、降低IgE介导的I型变态反应相关。

[Abstract] Objective To investigate the effect of capsaicin on nasal mucosal morphology and substance P expression in allergic rhinitis rats and its mechanism. Methods Animal model of allergic rhinitis was established. Capsaicin was used intranasally, and the number of sneezes and catching nose, runny nose situation were recorded and scored, and the scores were superimposed. The nasal mucosal morphology among normal group, untreated group and treated group was compared. The changes of nasal mucosa mast cells and substance P content after treatment were compared. Results The symptoms of allergic rhinitis in the treatment group after treatment were significantly relieved(P<0.05). The nasal mucosa mast cells and the content of substance P in the treatment group were significantly lower than those in the untreated group(P<0.05). Conclusion Capsaicin has a therapeutic effect on allergic rhinitis, and its mechanism is related to reduce the content of substance P and reducing IgE-mediated type Ⅰ allergic reaction.

近年来研究发现,变应性鼻炎(allergic rhinitis,AR)的发生与神经肽如P物质(SP)、降钙素基因相关肽(CGRP)关系密切[1]。这些神经肽与血流量和鼻黏膜腺体的分泌有关,并参与免疫应答的调节[2]。SP可以增强神经兴奋性,若SP大量释放,可引起鼻分泌的亢进[3-4]。辣椒素(capsaicin,CAP)可以选择性消耗神经末梢释放出的神经肽类物质,因此又被称为P物质耗竭剂[5-6],可以使感觉神经敏感性下降。本研究通过建立AR动物模型观察辣椒素对AR大鼠症状、鼻黏膜形态、P物质表达的影响,从而探讨辣椒素在变应性鼻炎防治中的作用机制。
1 材料与方法
1.1动物及分组
健康雄性SD大鼠[6]30只[动物批号:SCXK(京),2016-0002],体重180~220 g;饲养条件:宁夏医科大学动物实验中心,无特殊病原体(SPF);饲养湿度:40%,饲养温度:(22±2)℃。随机分为三组,每组10只,分别为正常对照组(正常组)、变应性鼻炎未治疗组(未治疗组)、变应性鼻炎辣椒素治疗组(治疗组)。应用苦杏仁酸进行标记,用以区别分组。
1.2 药物及试剂
卵清蛋白(Sigma公司生产)、氢氧化铝粉末(Sigma公司生产)、辣椒素(Sigma公司生产)、P物质抗体[艾博抗(上海)贸易有限公司]。……
