[摘要] 具有二芳基甲基哌嗪的化合物具有广泛的药理活性，包括抗组胺、抗肿瘤、抗高血压、抗偏头痛等，通过整理文献，本文对近年来发表的二芳基甲基哌嗪化合物的药理活性研究文献进行整理和分析，为深入开展以含有二芳基甲基哌嗪结构为创新新药研究提供信息。

Pharmacological activities and development of Diarylmethyl Piperazine derivatives

[Abstract] The structural fragment of Diarylmethyl Piperazine derivatives have multiple pharmacological activities， including antihistaminie， antitumor， antihypersive， antimigraine and so on. This article summarizes and analyzes the study literatures on the pharmacological activity of diarylmethyl piperazine derivatives published in recent years so as to provide information for studies on new drugs of diarylmethyl piperazine derivatives on the basis of the pharmacological activity.

哌嗪是许多重要药物的结构片断，在哌嗪的1、4位引入不同的取代基可产生多种不同的药理活性，二芳基甲基哌嗪就是在哌嗪的1位引入二芳基甲基，在4位引入其他取代基的。综合几年来的药物发现，具有二芳基甲基哌嗪结构的药物主要集中在抗组胺、抗肿瘤、抗高血压、抗偏头痛等方面[1]。本文将针对该类化合物的药理作用、结构特点、代表性药物做简要介绍。

1 抗炎及抗过敏药

抗炎及抗过敏药物是临床中广泛使用的品种，主要用于人体免疫系统变应性疾病的脱敏治疗。近年来，随着过敏性疾病发病率的不断上升，抗炎及抗过敏药物的用药量也呈增长趋势。在众多的抗炎及抗过敏药物中，其中相当一部分药物都具有二芳基甲基哌嗪结构[2-3]。见图1。

西替利嗪（Cetirizine）是美国辉瑞、比利时联合化工集团UCB公司开发的第二代抗组胺药物[4-5]。西替利嗪具有长效选择性抗H1受体的作用，且无镇静作用，一上市就取得了不错的销售业绩。在2003年全球市场突破20亿美元，是欧洲市场销售最多的抗过敏药物。……
