响应面法优化岭南炮天雄生物碱提取工艺

[摘要] 目的 优化岭南特色饮片炮天雄总生物碱的提取工艺。 方法 采用响应面分析法,依据Box-Behnken原理优化炮天雄乙醇超声提取工艺,实验以总生物碱提取率为评价指标,考察提取时间、乙醇浓度、液料比等主要影响因素,最终得出炮天雄最佳提取工艺。 结果 炮天雄总生物碱最佳提取工艺为乙醇比例75%,液料比8.0,提取时间30 min,在此条件下,总生物碱提取率理论值为31.1%,实际值为32.5%。 结论 基于响应面法优化的参数条件具有较高的实用价值,可为岭南特色炮天雄的开发利用提供参考依据。

[Abstract] Objective To optimize extraction process of total alkaloids from Lingnan featured pieces Paotianxiong by response surface method. Methods Three-variables as extraction time, ratio of material to solution and ethanol concentration, and three-levels Box-Benhnken experimental design was used to optimize the effects of independent variables on the total alkaloid extraction efficiency. Results The optimum conditions were extraction time 30 min, ratio of material to solution 8.0, and 75% ethanol concentration. Under the optimum conditions, the theoretical total alkaloid extraction rate was 31.1%, and the actual rate was 32.5%. Conclusion Response surface method is an effective methodology to optimize the total alkaloid extraction process of Lingnan featured pieces Paotianxiong.

附子为毛茛科植物乌头(Aconitum carmichaeli debx.)的子根,传统中医认为子根中独生个大者称之为天雄[1],岭南地区根据传统用药习惯,采用传统炮制技术将其加工成独具岭南特色炮制品,习称炮天雄[2]。炮天雄在岭南地区具有广泛临床应用基础,在港澳、东南亚地区,炮天雄常作为滋补食疗上品,用于治疗元阳素虚、肾亏阳虚证患者[3-5]。现代研究表明,炮天雄富含生物碱类成分[6-8],这类成分为炮天雄的主要药效成分,因此从炮天雄分离提取生物碱类成分具有较高的现实意义。目前,嶺南特色炮天雄生物碱成分提取工艺的研究未见报道。响应面法(RSM)是一种优化多变量体系的有效实验工具,采用多元二次回归方程来拟合多个因素与响应值之间、因素与因素之间的函数关系,找出各因素的最佳组合和响应值的最优值,有效地解决多变量问题[9-12]。……
