微流控芯片快速测定三颗针与盐酸小檗碱片中小檗碱

[摘要] 目的 建立了微流控芯片非接触电导检测法测定三颗针与盐酸小檗碱片中小檗碱的分析方法。 方法 选择5 mmol/L 乙二胺 + 15 mmol/L H3BO3作缓冲溶液;加入0.4 mmol/L β-环糊精(β-CD) 添加剂;分离电压2.50 kV;进样时间10 s; 结果 盐酸小檗碱的线性范围为10~4.0×102 μg/mL(r = 0.997),检出限为10 μg/mL(S/N = 3),三颗针中小檗碱平均含量为0.14%,盐酸小檗碱片中小檗碱平均含量为91.6 mg/片,其平均标准偏差(RSD)分别为1.3%(n = 6)、1.4%(n = 6)。平均加标回收率分别为103%和98.9%。 结论 该方法快速、简单、高效,可为三颗针及盐酸小檗碱片提供新的分析方法。

Determination of Berberine in Barberry Root and Berberine hydrochloride Tablets by Microfluidic Chip

[Abstract] Objective To establish a new method for the determination of Berberine in Barberry Root and Berberine hydrochloride Tablets by microfluidic chip with contactless conductivity detection. Methods 5 mmol/L ethylene diamine + 15 mmol/L boric acid + 0.4 mmol/L β-CD were selected as buffer solution, 2.50 kV was chosen as separation voltage with injecting time of 10 s. Results The linear range of berberine ranged from 10 to 4.0×102 μg/mL (r = 0.997) with the detection limit of 10 μg/mL (S/N = 3), the average recovery was 103% and 98.9% respectively. The average concentration of berberine in barberry root was 0.14% (RSD = 1.3%, n = 6) and the average content of berberine in berberine hydrochloride tablet was 91.6 mg (RSD = 1.4%, n = 6). Conclusion The method is simple, rapid and efficient, which can provide a new analysis method for Berberine and Barberry root.

三颗针为小檗科(Berberidaceae)小檗属(Beris)植物的俗称,其含有的生物碱主要有小檗碱,具有清热解毒、抗菌等作用[1]。盐酸小檗碱片常用于治疗肠道感染。其分析方法包括高效液相色譜法[1-7]、紫外分光光度法[8-9]、薄层扫描法[10-11]和毛细管电泳法[12-15]等。高效液相色谱分离效率较高、定量效果好、分析速度较快,但需分离柱、高压泵及较大量的流动相,分析成本较高,且流动相多为有毒溶剂,对环境有一定污染;紫外分光光度法易受共存成分的干扰;相比而言,微流控芯片分析技术所需样品量及缓冲液量少,分析耗费低,环境污染少,样品预处理简单[16-18]。可为三颗针与盐酸小檗碱片的质量控制提供一种新的分析方法。
1 材料与方法
