【摘 要】 目的：建立在消栓颗粒剂中有效成份黄芪甲苷的HPLC-ELSD含量测定方法。方法：用Nanologica SVEA C18色谱柱（250mm×4.6mm，5m）采用HPLC-ELSD法测定黄芪甲苷，蒸发光散射检测器（SEDEX-55型），柱温箱为25℃，流动相为乙腈∶[KG-*3/5]水（35∶[KG-*3/5]65），流速為1.0mL/min。结果：黄芪甲苷在0.2742～1.1492g内有良好的线性关系R=0.9994（n=6），平均回收率为99.16%，RSD为1.21%。结论：本方法操作简单快捷，结果准确可靠，重复性好，可用于消栓颗粒剂的质量控制。

Abstract：Objective To establish an HPLC-ELSD method for determining the effective component of astragaloside A in Xiaoshuan Granules. MethodNanologica SVEA C18 column （4.6mm×250mm， 5m） HPLC-ELSD determination of astragaloside， SEDEX-55 type evaporative light scattering detector， column temperature box is 25℃， mobile phase is acetonitrile： water （35∶[KG-*3/5]65 ）， Flow rate∶[KG-*3/5]1.0mL/ min. ResultsAstragaloside A is in the range of 0.2742～1.1492g， the linear relationship between the injection volume and the peak area integral value is good， the average recovery rate of R2 =0.9994 （n=6） is 99.16%， and the RSD is 1.21%.ConclusionThe method is simple， accurate， reliable and reproducible for the quality control of Xiaoshuan granule.

消栓颗粒剂出自清朝名医王清任的《医林改错》卷下瘫痿论方[1]。该方特点为重用黄芪为君药，其黄芪用量是其它活血药总和的约3倍，方中重用的黄芪中含有多糖及黄芪甲苷[2-4]，同时黄芪还具有抗炎、增强免疫[5-6]、抗氧化[7]、保护心脏[8]、调脂[9]、降血糖[10]等作用。黄芪甲苷作为黄芪的主要有效成分，在我国药典中有含量测定并有相关含量限度规定[11]。本研究建立消栓颗粒剂中黄芪甲苷含量测定方法，进一步细化药品的质量控制。

1 仪器与试药

1.1 实验仪器 液相色谱仪（Agilent 1100） 检测器为四元泵二极管阵列，数据转换器为Agilent35900E蒸发散射检测器（SEDEX-55），单元泵（DXNamax-SD-200） ，AgII空气发生器电子天平（Mettler instrument Corp），超声波提取器（型号GT-350W型济宁科特超声电子有限公司）。

1.2 药品 消栓颗粒（ 自制的三批药品分别为20190905 20190904 20190902）自制的黄芪阴性对照;黄芪甲苷对照品（中检所110781201717）;乙腈（色谱纯）;其它试剂（分析纯）。

2 方法与结果

2.1 溶液的制备 ①供试品溶液的制备：取本品7g，精密称定，加氨试液40mL移至具塞三角瓶中摇匀，密闭，过夜。……
