白细胞介素-6启动子区rs200852701(G/A)与南通地区汉族人群脓毒症发病关系的研究

【摘要】 目的 探讨江苏南通地区汉族人群中白细胞介素-6(IL-6)基因3′UTR的rs200852701(G/A)多态位点单核苷酸多态性(SNP)与脓毒症发病之间的关系。方法 51例脓毒症患者作为实验组, 领选择52例同期门诊行健康体检患者作为对照组。分别提取实验组和对照组基因组DNA, 聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术扩增目的基因片段并测序。分析IL-6基因启动子区rs200852701(G/A)与脓毒症发病之间的关系。结果 对本实验的IL-6基因聚合酶链式反应(PCR)产物, 进行相关基因分型, 结果显示, 研究对象中IL-6的SNP分布符合Hardy-Weinberg遗传平衡。测序结果表明IL-6基因存在SNP位点rs200852701(G/A)。实验组的血浆IL-6水平为(14.23±6.1)pg/ml, 高于对照组的(11.07±3.81)pg/ml, 差异有统计学意义(P<0.05)。结论 IL-6基因3′UTR的靶位点rs200852701(G/A)多态位点SNP与脓毒症发病之间存在一定关系。

【Abstract】 Objective   To discuss the correlation between single nucleotide polymorphism (SNP) of interleukin-6 (IL-6) gene 3 'UTR rs200852701 (G/A) and sepsis in Han population in Nantong, Jiangsu province. Methods   There was 51 sepsis patients as the experimental group, and concurrent 52 healthy examinee as the control group. Genomic DNA was extracted from sepsis group and control group, and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technology was used to amplify and sequence the target gene. The correlation between IL-6 gene promoter rs200852701 (G/A) and sepsis was analyzed. Results   The IL-6 gene polymerase chain reaction (PCR) product of this experiment was subjected to related genotyping. The results showed that the distribution of SNP of IL-6 in the study subjects was in line with Hardy-Weinberg genetic balance. Sequencing results showed that there was a SNP site rs200852701 (G/A) in the IL-6 gene. The plasma IL-6 of the experimental group was (14.23±6.1) pg/ml, which was higher than that of the control group (11.07±3.81) pg/ml, and the difference was statistically significant (P<0.05). Conclusion   IL-6 gene 3′UTR rs200852701 (G/A) polymorphism site is associated with sepsis.

膿毒症是因感染导致的系统性炎症反应, 可以导致多器官功能衰竭和死亡。也是目前医院患者死亡率和医疗资源消耗的主要严重疾病之一。在美国, 严重脓毒症占重症监护室(ICU)患者发病率的10.00%, 致死率可高达50.00%[1]。而德国2013年有280000例住院患者被诊断为脓毒症, 其中7500例患者死亡[2]。近年来, 控制脓毒症的发病率和死亡率一直是各国重点研究的领域之一。随着认识的深入, 目前脓毒症的发病率和死亡率较以往都有了一定程度下降[3]。IL-6是参与系统炎症反应的促炎细胞因子之一。先天性和获得性免疫反应中起着重要作用。它促进炎症现象, 比如刺激淋巴细胞的增殖, 促进B细胞的分化, 是一种最重要的炎症急性期的反应介质[4]。IL-6由多种类型的免疫细胞产生, 包括激活巨噬细胞、单核细胞和淋巴细胞。……
