摘要 目的：利用单侧输尿管梗阻模型小鼠筛选糖肾方颗粒的最佳制备工艺。方法：根据大孔树脂富集、醇提、水提方法设计糖肾方制备工艺1、2、3组。选用C57BL/6小鼠，随机分为假手术组，模型组，工艺路线1、2、3组，阳性药福辛普利组，连续给药6d后，进行单侧输尿管结扎手术。手术后再连续给药7d，检测小鼠血清肌酐和尿素氮，根据MASSON染色半定量分析各组小鼠的肾间质纤维化，根据HE染色进行肾间质细胞浸润评分。结果：各工艺路线组均对UUO引起的小鼠肾病理损伤有一定的改善作用，均显著改善肾间质纤维化，差异有统计学意义（P<0.05），其中工艺路线1、3组作用更佳。工艺路线3组能显著改善小鼠UUO后的血肌酐和尿素氮水平，显著减少肾间质细胞浸润，差异有统计学意义（P<0.05）。结论：糖肾方颗粒的制备工艺确定为水提工艺。

AbstractObjective：Toselecttheoptimalpreparationprocessofsugar-basedkidneygranulesinunilateralureteralobstructionmodelmice.Methods：Accordingtomacroporousresinenrichment，alcoholextractionandwaterextractionmethods，thepreparationprocessofTangshenFormulaI，IIandIIIweredesigned.C57BL/6micewererandomlydividedintoshamoperationgroup，modelgroup，routeI，IIandIIIgroups，andpositivedrugfosinoprilgroup.Sixdaysaftercontinuousadministration，unilateralureteralligationwasperformed.Sevendaysaftersurgery，miceweregivenserumcreatinineandureanitrogenfor7days.SemiquantitativeanalysisofrenalinterstitialfibrosisineachgroupwasperformedaccordingtoMASSONstaining.RenalinterstitialcellinfiltrationscoreswereevaluatedbyHEstaining.Results：EachroutegrouphadacertainimprovementonthepathologicaldamageofkidneyinducedbyUUOinmice，allofwhichsignificantlyimprovedrenalinterstitialfibrosis（P<0.05）.Amongthem，thefirstandthirdgroupsoftheprocessroutehadbettereffects.ThethreeroutescansignificantlyimprovetheserumcreatinineandureanitrogenlevelsofmiceafterUUO，andsignificantlyreducerenalinterstitialcellinfiltration（P<0.05）.Conclusion：ThepreparationprocessofTangshenFormulawasdeterminedasthewaterextractionprocess.

肾间质纤维化（RenalInterstitialFibrosis，RIF）是许多类型肾脏疾病的主要病理基础，造成肾脏功能的不可逆损伤，同时在糖尿病肾病中肾间质纤维化促进病情进展，因此抑制RIF是保护肾脏的有效治疗策略。中医药防治肾间质纤维化有其独特的优势。糖肾方（TangShenFomula，TSF）是本课题组继承前人经验，结合临床实践，以益气养阴、活血通络立法，组方而来。它是由黄芪，生地黄、山茱萸、三七、卫矛、熟大黄、枳壳七味中药组成的复方制剂。临床试验和基础研究证明TSF可以较好的防治糖尿病肾病和改善肾间质纤维化[1-2]。本研究利用经典的单侧输尿管梗阻（UnilateralUreteralObstruct，UUO）小鼠动物模型，对大孔树脂富集、醇提以及水提工艺制备的糖肾方颗粒进行评价，以确定糖肾方最优提取方法。……
