【摘 要】 目的：通过研究油桐花水提物外用对烧伤大鼠的影响，并探索其对Bcl-2、Bax基因表达的影响。方法：60只Wistar大鼠，随机分为6组，每组10只。除正常组外其余5组建立大鼠深Ⅱ度烧伤模型，并对其进行相应药物治疗。待伤势恢复后，相机拍摄各组患处，处理取材。用Elisa和免疫组化法检测Bcl-2和Bax的表达情况。结果：Elisa检测表明，与模型组比较，各用药组血清中Bcl-2表达明显上调（P<0.01）；HE染色结果显示，模型组有大量纤维组织增生及炎性细胞浸润，各用药组炎性细胞浸润缓解；免疫组化实验结果显示，与模型组比较，各用药组与正常组促凋亡因子Bax表达水平比较明显降低；各治疗组中抗凋亡因子Bcl-2表达水平均有上调。结论：油桐花水煎剂对大鼠烧伤有明显治疗作用，与阳性药物烧烫伤膏作用相似，可抑制细胞凋亡，有利于恢复烧烫伤创面。

Abstract：Objective To study the effect of external application of water extract of Tung Flower flower on burned rats and explore its influence on the expression of Bcl-2 and Bax gene. Methods Sixty Wistar rats were randomly divided into 6 groups of 10 each. Rats with deep second degree burns were established in the other 5 groups except the normal group and treated with corresponding drugs. After the injury recovers， the camera photographs the affected area and processes the materials. The expression of Bcl-2 and Bax was detected by Elisa and immunohistochemistry. Results Elisa test showed that the expression of Bcl-2 in the serum of each drug group was significantly increased compared with the model group （P<0.01）. The results of HE staining showed that the model group had a large number of fibrous tissue proliferation and inflammatory cell infiltration， and the inflammatory cell infiltration in each drug group relieved. The results of immunohistochemistry showed that compared with the model group， the expression level of pro-apoptosis factor Bax was significantly decreased in each drug group and normal group； the expression level of anti-apoptosis factor Bcl-2 was up-regulated in each treatment group. Conclusion Tung oil flower decoction has obvious curative effect on burns in rats. Similar to the burn and burn cream on the market， it can inhibit cell apoptosis and is beneficial to the recovery of burn wounds.

中医认为烧烫伤属于“烫火伤”范畴，常因外袭火毒，灼溃皮肤，皮肉焦卷，疼痛难忍甚至伤筋动骨，有些患者因火毒内攻，出现较重的全身症状[1]。油桐花具有消肿、镇痛、止血、生肌、促进伤口愈合等作用[2]，在烧烫伤治疗方面，有很好的疗效[3-4]。作为凋亡家族中最主要成员，Bcl-2与Bax在烧伤中也有重要作用[5-6]。但目前未见桐子花水提物调节Bcl-2与Bax表达干预烧烫伤相关文献报道。实验拟通过油桐花水提液调节凋亡蛋白Bcl-2、Bax干预烧烫伤，初步确定油桐花对烧烫伤的作用机制。

1 实验材料

1.1 实验动物 SPF级Wistar雄性大鼠60只，体重（150±20）g，适应性喂养一周，购于长沙市天勤生物技术有限公司，动物许可证编号：SCXK（湘）2014-0011。

1.2 药品与试剂 油桐花采自贵州省遵义市正安县并由贵阳中医学院孙庆文教授鉴定，无水乙醇（批号：20150601，重庆川东化工集团有限公司），二甲苯（批号：20150508，重庆茂业化学试剂有限公司），烧烫伤膏（批号：20150601，长春海外制药集团有限公司），苏木素染色液（批号：AR1180-1，博士德生物有限公司），伊紅染色液（批号：AR1180-2，博士德生物有限公司），Elisa检测试剂盒（批号：CK-E30187R，上海研辉生物科技有限公司）。……
