摘要[目的]优化提取枸杞总黄酮的最佳工艺条件。 [方法]以宁夏枸杞作为研究对象，采用回流-超声复合式提取方法，分别通过单因素试验及正交试验设计，对枸杞中的有效成分总黄酮的提取和测定工艺条件进行优化。 [结果]得到枸杞中总黄酮测定的最优条件为 利用石油醚对枸杞先进行回流提取，然后使用60%的乙醇溶液，料液比为1∶40 （g∶mL），在70 ℃条件下提取30 min，取次数为2次。利用优化参数对枸杞中总黄酮的含量测定，经计算，该方法的相对标准偏差（RSD）为0.91%，加标回收率值在98%以上。 [结论]该方法具有准确率高、成本低、设备简单、操作便捷、易于推广的特点。

Study on the Optimized Conditions by Refluxultrasonic Extraction Flavones from Lycium barbarum

Abstract[Objective]To optimize the extraction technology of total flavonoids from Lycium barbarum L. [Method]Taking Ningxia Lycium barbarum L. as the research object， the method of reflux ultrasonic combined extraction was adopted.The extraction and determination of total flavonoids in Lycium barbarum L. were optimized by single factor test and orthogonal design.[Result]The optimum conditions for the determination of total flavonoids in Lycium barbarum were as follows： petroleum ether was used to extract the total flavonoids from Lycium barbarum by reflux， then 60% ethanol solution was used， the ratio of material to liquid was 1∶40 （g∶mL）， and the extraction time was 2 times at 70 ℃ for 30 minutes.The content of total flavonoids in Lycium barbarum was determined by the optimized parameters. The relative standard deviation （RSD） of the method was 0.91%， and the recovery was over 98%.[Conclusion] The method has the characteristics of high accuracy， low cost， simple equipment， convenient operation and easy popularization.

枸杞（Lycium chinense）別名苟起子、甜菜子、血枸子，属于茄科枸杞属植物，国内主要产于宁夏、新疆、甘肃等地区，其中属宁夏地区产出的枸杞最为有名[1]。枸杞具有较高的药用价值，具有解热及治疗糖尿病、高血压、肝肾疾病等功效[2-5]。通过对枸杞有效成分研究发现，枸杞中含有较高的黄酮及芦丁，其中黄酮类化合物具有多种生物活性，具有抗氧化性和降血脂作用，对预防动脉粥样硬化有较好的疗效[6]。因此，如何高效提取枸杞叶中总黄酮并进行测量成为人们关注的焦点。笔者采用回流-超声辅助复合提取法对枸杞中的有效成分总黄酮进行了提取，通过对提取条件进行单因素试验和正交试验，进一步优化提取枸杞总黄酮的最佳工艺条件，并采用NaNO2-Al（NO3）3-NaOH分光光度比色法，对枸杞中总黄酮的含量进行测试。……
