甘草黄酮对UVB损伤HDF细胞保护作用研究

摘要[目的]研究甘草黄酮对UVB照射后产生光老化现象的HDF细胞保护作用。[方法]采用30 mJ/cm2的UVB照射HDF细胞建立损伤模型;加入不同浓度的甘草黄酮,24 h后MTT法检测细胞活性;用试剂盒法测定细胞上清液中谷胱甘肽过氧化物酶(GSHPx)、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量。[结果]甘草黄酮浓度为10-9、10-10、10-11 mol/L时,能使受UVB照射损伤的HDF细胞活性增强;甘草黄酮浓度为10-9、10-10 mol/L时,GSHPx、SOD活性升高,MDA含量降低;甘草黄酮浓度为10-11 mol/L時,SOD活性升高、MDA含量降低。[结论]甘草黄酮能减轻UVB对HDF细胞的损伤,其机制可能与抑制氧化损伤、提高SOD活性有关。

Effects of Licoflavone on HDF Cytoprotection after UVB Injury

Abstract[Objective] To research the effects of licoflavone on HDF cytoprotection after UVB injury. [Method] HDF was irradiated under 30 mJ/cm2 UVB to establish injury mode. After adding different concentrations of licoflavone, cell viability was detected by MTT method after 24 h. Contents of GSHPx, SOD and MDA in cell supernatant were detected by kit method. [Result] When licoflavone concentration was 10-9, 10-10 and 10-11 mol/L, HDF activities enhanced after UVB injury. When licoflavone content was 10-9 and 10-10 mol/L, contents of GSHPx, SOD and MDA reduced. When licoflavone content was 10-11 mol/L, SOD activity enhanced and MDA content reduced. [Conclusion] Licoflavone reduces the HDF injury induced by UVB. Its mechanism might be related to the restriction of oxidative damage and the enhancement of SOD.

皮肤是身体与外界环境直接接触的第一道防线,长期暴露于紫外线下会产生氧化应激效应[1]。因紫外线中UVB极大部分被皮肤表皮所吸收,很难渗入皮肤内部,所以多数研究者选择研究UVB对于皮肤表皮层中角质细胞的作用。但由于UVB阶能较高,可到达真皮层的成纤维细胞(HDF),导致被照射部位细胞发生代谢反应,伴随产生活性氧簇(ROS)[2],抗氧化防御系统被破坏,产生机体氧化损伤,直接表现为皮肤松弛、干燥、纹理变粗、癌变等[3]。在抑制ROS诱导的损伤方面,人体内存在的多种抗氧化酶和非酶性抗氧化物发挥着巨大的作用,谷胱甘肽过氧化物酶(Glutathione peroxidase,GSHPx)、超氧化物歧化酶(SOD)等抗氧化酶可以清除机体内产生的ROS,维持机体平衡,但当机体内抗氧化物酶的活性降低或ROS产生过量时,机体细胞会受到损伤,引起氧化-抗氧化平衡系统破坏,产生DNA损伤、酶活性丧失及细胞凋亡等,发生皮肤发炎、皱纹,机体出现疾病(肿瘤)等[4]。……
