受精液种类、卵丘细胞及卵巢保存温度对奶牛体外受精效果的影响

摘 要:为了提高奶牛体外受精效果,研究了体外成熟卵母细胞在IVF-100、BO和TALP三种体外受精液中的受精效果,同时比较了裸卵(NOs)与卵丘卵母细胞复合体(COCs)对受精效果的影响,卵巢的不同保存温度对卵母细胞受精效果的影响。结果显示,体外成熟卵母细胞在受精液IVF-100中的卵裂率显著高于BO液的(P<0.05),而TALP液中的卵裂率与IVF-100和BO液的差异都不显著(P>0.05);裸卵(NOs)的卵裂率低于卵丘卵母细胞复合体(COCs),但差异不显著(P>0.05);NOs的囊胚率极显著低于COCs的(P<0.01);卵巢在10和37 ℃下保存2~3 h,卵母细胞卵裂率和囊胚率差异均不显著(P>0.05)。

Abstract: In order to improve the effect of fertilization in vitro of cows, this study compared the effect of fertilization among the IVF-100, BO and TALP solution. Meanwhile, this test compared the effect of fertilization between NOs and COCs,and the trail explored the difference of effect of fertilization on different temperature for ovaries. The results showed that the rate of cleavages of the oocytes in IVF-100 was significantly higher than that in BO (P<0.05),while the oocytes rate of cleavages of TALP solution had no significant differences with that in IVF-100 and BO (P>0.05).The rate of cleavages of NOs was lower than that of COCs, while there was no significant difference (P>0.05),but the NOs rate of blastocyst was extreme significantly lower than that of COCs (P<0.01). Also, there was no significant difference between the rate of cleavages and the rate of blastocyst when the ovaries stored in 10 and 37 ℃ for 2~3 hours (P>0.05).

随着配子与胚胎生物技术的研究与应用,对胚胎的需求量越来越大。超数排卵和胚胎移植的合理化应用虽能成倍提高优良母畜的繁殖率,但所获得的胚胎数量有限。为了解决这个问题,近年来国内外对奶牛卵母细胞体外受精技术进行了大量的研究。保国俊、Ginther等[1-2]通过IVF技术可提供数量更多、成本更低的胚胎,体外受精技术是对优良公、母畜的综合充分利用。近年来,牛体外受精技术 (In vitro fertilifation, IVF)取得了较大进展,已成为可在实验室条件下生产大量廉价胚胎的实用技术,将为胚胎移植提供重要的胚胎来源。牛体外受精技术一直是研究的热点。但由于体外受精技术操作环节复杂,操作过程涉及因素较多以及卵母细胞对体外生存环境要求较高,使得试验体系不稳定,进而影响试验结果的准确性[3]。

本试验通过对比不同受精液对卵母细胞体外受精效果影响的研究,分析了不同形态的卵子和卵巢运输保存温度对奶牛成熟卵母细胞体外受精能力的影响,皆在为优化奶牛体外受精技术奠定基础。
