甲基化衍生—高效液相色谱法检测北五味子中的代森锰锌

摘要:利用甲基化衍生-高效液相色谱法检测北五味子[Schisandra chinensis(Turcz.)Bail.]中代森锰锌残留量。以BEH C18(2.1 mm×50 mm,1.7 μm)为固定相;乙腈∶水(18∶82,V/V)为流动相;检测波长为 272 nm。结果表明,代森锰锌的平均回收率达97.0%,RSD为1.36%,最低检出限(LOD)为0.02 mg/kg,最低检测浓度(LOQ)为0.05 mg/kg,结果符合农药残留试验准则要求。北五味子样品中均未检出代森锰锌。

Abstract: A methylation-derived UPLC for detecting mancozeb residue in Schisandra chinensis(Turcz.)Bail. was studied in this paper. The analysis was performed on the column of BEH C18(2.1 mm×50 mm,1.7 μm) with the mobile phase of acetonitrile-water(18∶82,V/V), the wavelenth detected was 272 nm. The results showed that the average recovery reached 97.0% with RSD of 1.36%. The LOD and LOQ were 0.02 and 0.05 mg/kg,respectively. This satisfied results were fit for the testing standard of pesticide residue.No mancozeb residues was detected in the samples.

代森锰锌(Mancozed)属乙撑二硫代氨基甲酸酯(盐)类杀菌剂(EBDCs),是锌离子与代森锰的配位络合物,分子式(C4H6MnN2S4)xZny(x∶y=1∶0.091,MW≈270),溶解性差,不溶于水和常见有机溶剂,具有广谱低毒低残留高效力特性,对北五味子[Schisandra chinensis(Turcz.)Bail.]黑斑病具有良好的防治效果[1]。EBDCs本身无食品安全风险,但是在生产、贮存和使用过程中降解产生的主要代谢产物乙撑硫脲(ETU)却具有致癌、致畸、致突变作用,危害人体健康。北五味子为传统中药,出口韩国、日本及东南亚地区,检测代森锰锌的残留量对提高我国北五味子产值具有重要意义。
目前代森锰锌测定主要包括分光光度法、毛细管电泳法、顶空气相色谱法和甲基化衍生液相色谱法等[2]。其中分光光度法和毛细管电泳法灵敏度低,仅能检测高浓度样品;顶空气相色谱法通常被列为各国标准分析方法,但该法分析的是EBDCs类杀菌剂产生的二硫化碳总量,缺乏专一性;甲基化衍生原理最早被Gustafsson和Thompson应用于福美双的含量分析,马婧玮和潘灿平等人在其基础上建立起代森锰锌的分析方法,其原理为在碱性条件下,用乙二胺四乙酸二钠(EDTA)将代森锰锌转化为代森钠,代森钠能与碘甲烷发生甲基化反应,得到甲基化产物,最后由HPLC检测甲基化产物。……
