蒙古口蘑多糖超声波辅助提取技术及含量测定

摘要:以蒙古口蘑(Tricholoma mongolicum imai)为研究对象,以多糖得率衡量超声波辅助提取工艺。在单因素试验基础上,采用L9(34)正交设计优化提取工艺。结果表明,影响口蘑多糖得率的因素依次为提取温度>盐酸浓度>提取时间>料液比,最佳提取工艺为0.2 mol/L盐酸、料液比1∶40(g∶mL),50 ℃超声波提取20 min。在此提取条件下,口蘑多糖的得率为(16.65±0.29)%,该提取方法简便、快速,可以为蒙古口蘑的质量控制和合理开发提供参考依据。

Optimization of Ultrasound-assisted Extraction and Content Determination of Polysaccharide from Tricholoma mongolicum Imai

Abstract:To optimize the ultrasound-assisted extraction process and content determination of polysaccharide from Tricholoma Mongolicum Imai, based on the single factor experiments, an orthogonal array was constructed to achieve the best conditions. The effects of influencing factors were extraction temperature> acid concentration >extraction time >solid-liquid ratio. The optimal conditions were as follows: extraction solvent 0.2 mol/L, solid-liquid ratio 1∶40, extraction time 20 min and extraction temperature 50 ℃. Under these optimal conditions, the polysaccharide yield was (16.65±0.29)%. This extraction method was simple and rapid, which could provide reference for the rational development and quality control of Tricholoma Mongolicum Imai.

蒙古口蘑(Tricholoma mongolicum imai)又称白蘑,为白蘑科白蘑属(口蘑属)真菌,主要分布于中国河北、内蒙古、黑龙江、吉林、辽宁等地区[1,2]。其菌肉肥厚、质地细嫩、味美独特,还可供药用,性平、味甘、宣肠益气、散热解表。植物多糖具有丰富的药理活性,如抗氧化、抗疲劳、降血糖、降血脂、抗肿瘤、免疫调节、抑制α-葡萄糖苷酶、稳定蛋白质等作用[3-10],因此其研究越来越受到学者的关注。本试验以去离子水为介质,采用超声波辅助提取技术提取蒙古口蘑多糖,通过正交试验优化提取工艺,可为其工业化利用提供参考。

1 材料与方法

1.1 材料与仪器

蒙古口蘑产于河北省张家口市张北草原,干燥粉碎后过60目筛,密封保存备用。D-葡萄糖购于美国Fisher公司,浓硫酸、苯酚、乙醇均为分析纯,水为去离子水。

KQ-250DA型超声波清洗器(昆山市超声仪器有限公司)、紫外分光光度计(北京普析通用仪器有限责任公司)、Alc-210型电子天平(赛多利斯集团)。
