桃金娘根中没食子酸和总多酚含量测定

[摘要] 目的 测定桃金娘根中没食子酸和总多酚的含量。 方法 没食子酸的含量测定采用超高效液相色谱法。色谱条件:以甲醇-0.5%磷酸(15︰85)为流动相,采用二级管阵列检测器(PDA)检测,检测波长为215 nm,流速为0.2 mL/min。总多酚的含量测定采用分光光度法,以没食子酸为对照品,福林酚试剂为显色剂,在748 nm波长下进行测定。 结果 没食子酸在3.853~115.6 μg/mL(r = 0.9999)浓度范围内与峰面积呈良好的线性关系,回收率为98.73%~100.76%。总多酚分析的线性范围为1.0~7.0 μg/mL(r = 0.9995),回收率为98.04%~101.17%。 结论 该方法简便、快速、重现性好,可用于桃金娘根的质量控制及定量分析。

Determination of gallic acid and total polyphenol in Radix Rhodomyrti

[Abstract] Objective To determine the content of gallic acid and total polyphenol in Radix Rhodomyrti. Methods Gallic acid in Radix Rhodomyrti was determined by ultra performance liquid chromatography (UPLC) with photo-diode-array (PDA) at 215 nm, and the mobile phase was methol-0.5% phosphate acid solustion (15∶85) at a flow rate of 0.2 mL/min. The content of total polyphenol was analyzed by spectrophotometry at 748 nm, gallic acid was selected as the reference substance. The colour-developing agent was Folin-Ciocalteu′s phenol reagent. Results The linear range for gallic acid and total polyphenol was 3.853-115.6 μg/mL (r = 0.9999) and 1.0-7.0 μg/mL (r = 0.9995) respectively. The recovery rate of gallic acid was 98.73%-100.76% and recovery rate of total polyphenol was 98.04%-101.17%. Conclusion The method is simple, rapid and good reproducible, can be used for quality control and quantitative analysis of Radix Rhodomyrti.

桃金娘[Rhodomyrtus tomentosa(Ait.) Hassk]是一种优良的野生植物资源,尤盛产于中国和日本的南部等地区[1-2];桃金娘根(Radix Rhodomyrti)为桃金娘的干燥根,含有黄酮苷、花色苷等黄酮类成分以及植物多糖成分和没食子酸等化合物多种活性成分[3-5],以“通经活血、收敛止血”功效收载于《兽药国家标准(化学药品、中药卷)第一册》中[6]。没食子酸是可水解鞣质的组成部分[7-8],具有抗感染、抗突变、抗氧化等多种生物学活性[9-10]。
桃金娘根药材及饮片质量标准首次收载于农业部《兽药规范》1992版二部[11],并未收入以后各版《中国兽药典》,该质量标准一直沿用至今[12]。该质量标准相对比较简单,专属性不强,也不够全面,未对其所含特征性生物活性成分进行色谱法鉴别,也未建立科学、准确、灵敏的光谱,色谱及质谱等方法对特征化学成分进行含量测定。……
