盐酸甲氧氯普胺注射液中抗氧剂亚硫酸氢钠的含量测定

摘 要 目的:建立盐酸甲氧氯普胺注射液中抗氧剂亚硫酸氢钠的含量测定方法。方法:基于在SO2存在下能使酸性品红溶液褪色的原理,采用比色法在549 nm 波长处测定吸光度, 用标准曲线法测定含量。结果:无水亚硫酸钠在9.917~29.75 mg/ml,吸光度的倒数与其浓度(以SO2计)呈良好的线性关系,r = 0.999,平均回收率为99.3%。结论:该方法操作简单,结果准确可靠,可作为制剂中抗氧剂亚硫酸氢钠的含量测定。

Determination of antioxidant sodium bisulfite
in metoclopramide dihydrochloride injection

ABSTRACT Objective: To develop a method for the determination of sodium bisulfite in metoclopramide dihydrochloride injection. Methods: Based on the fading of fuchsin acid solution in the present of SO2, a colorimetric method was established to be used for the determination of absorbance at the wavelength of 549 nm. The amount of sodium bisulfite in sample was calculated by reference to a calibration curve obtained. Results: The reciprocal of absorbance and the concentrations of anhydrous sodium sulfite (calculated based on sulfur dioxide) showed a good linear relationship over the range of 9.917~29.75 mg/ml (r=0.999) with an average recovery of 99.3%. Conclusion: This method is simple and accurate and can be used for the determination of sodium bisulfite in metoclopramide dihydrochloride injection.

甲氧氯普胺又名胃复安,是多巴胺受体阻断剂,可抑制胃平滑肌松弛,具有强大的中枢性镇吐作用,临床上普遍用于癌症放、化疗和手术后的呕吐、反流性食管炎、胆汁反流性胃炎等胃肠道疾病[1]。盐酸甲氧氯普胺注射液中常加入亚硫酸氢钠作为抗氧剂,目前该注射液现行国家标准尚未对抗氧剂的含量进行控制[2],也未有相关报道。亚硫酸氢钠在中国药典现行版中有收载,含量测定方法为容量分析法[2]1193,但该法不利于微量亚硫酸氢钠的测定,也无法消除注射液中其他成分的干扰。亚硫酸氢钠为亚硫酸氢钠与焦亚硫酸钠的混合物[2]1193,以二氧化硫(SO2)计算,文献[3-6]报道多采用离子色谱法和气相色谱法进行测定,考虑到成本问题,为增强注射剂的安全性,本文建立了比色法测定盐酸甲氧氯普胺注射液中亚硫酸氢钠的含量的方法。
