丹参饮介导过氧化物酶体增殖物激活型受体γ减轻大鼠冠脉粥样硬化

摘要 目的:研究丹參饮通过介导过氧化物酶体增殖物激活型受体γ(PPAR-γ)/肝X受体α(LXR-α)/三磷酸腺苷结合盒转运体A1(ABCA1)信号通路减轻大鼠冠状动脉粥样硬化的作用机制。方法:选取无特定病原体(SPF)级40只健康Wistar大鼠作为实验动物,随机分为空白组、冠状动脉粥样硬化组(对照组)、丹参饮低剂量观察组(低剂量组)和丹参饮高剂量观察组(高剂量组),比较各组大鼠的炎症介质水平、ICAM-1水平、脂代谢水平、PPAR-γ、LXR-α和ABCA1 mRNA表达和蛋白表达。结果:对照组、低剂量组和高剂量组的三酰甘油(TG)、胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)均高于空白组(P<0.05),低剂量组和高剂量组的TG、TC和LDL-C均低于对照组(P<0.05),且高剂量组低于低剂量组(P<0.05),但4组的HDL-C表达差异无统计学意义(P>0.05);对照组、低剂量组和高剂量组的炎症介质和ICAM-1表达均高于空白组(P<0.05),低剂量组和高剂量组的炎症介质和ICAM-1表达均低于对照组(P<0.05),且高剂量组低于低剂量组(P<0.05);对照组、低剂量组和高剂量组的PPAR-γ、LXR-α和ABCA1 mRNA和蛋白表达均高于空白组(P<0.05),且低剂量组和高剂量组明显高于对照组(P<0.05),高剂量组明显高于低剂量组(P<0.05)。结论:丹参饮可以通过对PPAR-γ-LXR-α-ABCA1信号通路的介导,发挥出抗冠状动脉粥样硬化的作用。

Abstract Objective:To study on Danshen Drink by mediating peroxisome proliferator-activated receptor γ(PPAR-γ)/liver X receptor α(LXR-α)/adenosine triphosphate binding cassette transporter A1(ABCA1) signaling pathway to alleviate the mechanism of coronary atherosclerosis of rats.Methods:A total of 40 healthy Wistar rats of SPF-free grade were selected as the experimental animals,and were randomly divided into a blank group,a coronary atherosclerosis group(control group),Danshen Drink low-dose treatment group(low-dose group) and Danshen Drink high Dose treatment group(high-dose group).The levels of inflammatory mediators,ICAM-1,lipid metabolism,PPAR-γ,LXR-α and ABCA1 mRNA expression and protein expression in each group of rats were compared.Results:The triacylglycerol(TG),cholesterol(TC),high-density lipoprotein cholesterol(HDL-C) and low-density lipoprotein cholesterol(LDL-C) of the control group,low-dose group and high-dose group were all higher than those of the blank group(P<0.05),the TG,TC and LDL-C of the low-dose group and the high-dose group were lower than those of the control group(P<0.05),and the high-dose group was lower than the low-dose group(P<0.05),but there was no statistically significant difference in the HDL-C expression of 4 groups(P>0.05); the expressions of inflammatory mediators and ICAM-1 in the control group,low-dose group and high-dose group were higher than those in the blank group(P<0.05),low-dose group and high-dose group The expressions of inflammatory mediators and ICAM-1 in the control group were lower than those in the control group(P<0.05).The protein expressions of PPAR-γ、LXR-α and ABCA1 mRNA were higher than those of the blank group(P<0.05).And the low-dose and high-dose groups were significantly higher than those of the control group(P<0.05),and the high-dose group was significantly higher than the low-dose group(P<0.05).Conclusion:Danshen Drink can play an anti-coronary atherosclerosis effect by mediating the PPAR-γ-LXR-α-ABCA1 signaling pathway.

冠状动脉是动脉粥样硬化的主要累及血管,冠状动脉粥样硬化会导致患者出现管腔闭塞或破裂、出血,增加了患者的心血管疾病发生率,威胁到了患者的身体健康[1-3]。近年来,相关临床研究发现过氧化物酶体增殖物激活型受体γ(PPAR-γ)/肝X受体α(LXR-α)/三磷酸腺苷结合盒转运体A1(ABCA1)信号通路与机体的动脉粥样硬化密切相关[4]。其中PPAR-γ的活化可以减少机体内趋化因子受体的表达,从而减少了斑块周围产生的炎症反应,发挥出了对动脉粥样硬化的预防作用[5-6]。基于此,本研究丹参饮通过介导PPAR-γ-LXR-α-ABCA1信号通路减轻大鼠冠状动脉粥样硬化的作用机制。……
