角质形成细胞生长因子促进大鼠皮肤伤口局部血管化效果研究

[摘要]目的:探索角质形成细胞生长因子(Keratinocyte growth factor,KGF)对于大鼠皮肤伤口局部的促血管化作用。以探究其可能的促进愈合的机理。方法:构建大鼠背部皮肤全层缺损模型,随机分为空白组(不作特殊处理),空白膜组(放置空白胶原膜),KGF膜组(放置加载KGF的胶原膜)。通过大体标本观察,组织学染色、免疫双标染色等技术对术后7d和14d时伤口的血管化程度进行比较和分析。结果:术后通过大体标本观察,可看到空白组和空白膜组虽均有血管长入伤口内,但中心部位血管化程度低于KGF膜组。Masson染色和免疫荧光染色及局部血管计数也证实加载KGF的胶原膜可以促进局部血管再生。结论:KGF可以促进大鼠皮肤伤口愈合过程中的血管化作用。

Keratinocyte Growth Factor Promotes Vascularization of Rat Skin Wounds

Abstract: Objective This article aims to explore the pro-angiogenic effect of KGF on rat skin wounds. Methods After successful modeling of full-thickness defect, 12 rats were randomly divided into three groups. The blank group was not treated. Blank collagen membrane was placed in the blank membrane group. Collagen membrane loaded with KGF was placed in the KGF membrane group. The degree of vascularization of the wounds at 7 and 14 days after surgery was compared and analyzed. Analysis was performed by gross specimen observation of wounds, histological staining, and immunolabeling. Results Through the observation of gross specimens, it can be seen that although wounds in the blank group and the blank membrane group have blood vessels that grow into the wound, the degree of vascularization in the central part was lower than that in the KGF membrane group. Masson staining and immunofluorescence staining also confirmed that collagen membranes with sustained release of KGF promoted local angiogenesis. Conclusion KGF can promote the vascularization of rat skin wound healing process.

發生在颜面部的皮肤损伤,如果发生延迟愈合或瘢痕愈合会很大程度影响患者的美观及功能。研究证实,血管化在皮肤愈合中起到十分关键的作用。如果缺少血管的滋养,新生上皮容易产生瘢痕,可能导致上皮化不完全,机械性能差等,因此血管化不足是决定和制约皮肤修复效果的关键因素之一[1-2]。研究发现,在皮肤受伤后,局部分泌多种细胞因子在真皮与表皮之间起重要的桥接作用,参与皮肤的愈合,改建及角化[3]。其中角质形成细胞生长因子(Keratinocyte growth factor,KGF)在正常皮肤中表达量较低,但皮肤受伤后KGF在短期内上调,提示其在伤口愈合和皮肤再生中起到关键作用。……
