正交试验法优化加味八珍合剂中特女贞苷的提取工艺

[摘要]目的 优化加味八珍合剂中特女贞苷的提取工艺。方法 采用正交试验法,以提取液中特女贞苷的含量为考察指标,对影响水煎提取效果的煎煮前浸渍时间(A)、加水倍数(B)、提取次数(C)与提取时间(D)四项作为考察因素进行研究,采用高效液相色谱法(HPLC)测定加味八珍合剂中特女贞苷的含量。结果 加味八珍合剂的最佳提取工艺为A2B1C2D1,即加6倍量的水,浸渍1 h,煎煮提取3次,每次1 h。结论 该实验所优选工艺得到的出膏率和特女贞苷的提出率最高。

Optimization of extraction process of Specnuezhenide in Jiawei Bazhen Mixture by orthogonal experiment

[Abstract] Objective To optimize the extraction process of Specnuezhenide in Jiawei Bazhen Mixture. Methods The orthogonal experiment method was used to determine the content of the Specnuezhenide in the extract as the index, and the immersion time (A), the water addition ratio (B), the extraction times (C) and the extraction time (D) as factors which affected the extraction effect of the decoction were studied. High performance liquid chromatography (HPLC) was used to determine the content of Specnuezhenide in Jiawei Bazhen Mixture. Results A2B1C2D1 was the best extraction process for Jiawei Bazhen Mixture, that was, adding 6 times of water, immersing for 1 hour, and boiling and extracting 3 times for 1 hour each time. Conclusion The preferred process for this experiment is to obtain the highest extraction rate of mixture and the highest extraction rate of Specnuezhenide.

加味八珍合劑作为南京中医药大学附属张家港医院(以下简称“我院”)的院内制剂,全方由多味中药组成,如茯苓、当归、川芎、女贞子等,具有益气血、补肝肾之效。加味八珍合剂是在八珍颗粒[1]的基础上增加了女贞子和狗脊两味中药,增强了滋肾阴、强腰膝之功效,主要用于气弱血亏、肾虚腰酸、四肢倦怠及妇女月经不调等症。方中女贞子具有扶正固本、滋补肝肾、明目乌发的功效[2]。随着当今研究的不断深入,女贞子更多的功效被挖掘出来,如在抗肿瘤、护肝[3]、延缓衰老、抗炎抗菌、调节免疫等方面都具有相应的疗效,并发现其对高血脂[4]、高血压、骨质疏松[5]等病症都有一定的效果[6]。现代药理研究表明女贞子的有效成分为特女贞苷[7],故本研究选择特女贞苷的含量作为水提正交工艺研究的评价指标,对……
