[摘要] 目的 探讨组蛋白去乙酰化酶抑制剂FK-228对Tca-8113细胞增殖、凋亡机制。 方法 低糖DMEM培养Tca-8113；倒置显微镜观察Tca-8113细胞受FK228的作用；在不同药物浓度处理24 h、48 h、72 h后利用CCK-8检测Tca-8113细胞增殖情况；Tca-8113凋亡细胞利用DNA ladde试剂盒检测。 结果 0.1 μg/mL，2 μg/mL，4 μg/mL FK-228对Tca-8113处理24 h、48 h、72 h后均有抑制作用，且与剂量、作用时间呈正相关。经FK228药物处理48 h后的细胞有DND LDEER出现。 结论 组蛋白去乙酰化酶抑制剂FK-228可呈降低人口腔鳞癌Tca-8113细胞活性，抑制其细胞增殖，诱导细胞凋亡。

Study on histone deacetylase inhibitor FK-228 in the induction of apoptosis of human tongue carcinoma cells

[Objective] Abstract To investigate the mechanism of proliferation and apoptosis of Tca-8113 cells by histone deacetylase inhibitor FK-228. Methods Tca-8113 was cultured by low glucose DMEM； the effect of FK228 on Tca-8113 cells was observed by inverted microscope； the proliferation of Tca-8113 cells was detected by CCK-8 at 24h， 48h and 72h after being treated by different drug concentrations. Tca-8113 apoptotic cells were detected by DNA ladde kit. Results After Tca-8113 was treated for 24 h， 48 h and 72 by 0.1 μg/mL， 2 μg/mL and 4 μg/mL FK-228， all showed an inhibitory effect， which was positively correlated with the dose and the time of effect. There were DND LDEER appeared in cells treated with FK228 for 48 hours. Conclusion The histone deacetylase inhibitor FK-228 reduces the activity of human oral squamous carcinoma Tca-8113 cells， inhibits cell proliferation and induces apoptosis.

口腔癌的發病率居全身恶性肿瘤的第六位，目前已是最常见的恶性肿瘤。口腔恶性肿瘤治疗不理想[1]，口腔鳞癌是头颈部常见恶性肿瘤，在我国其发病率占头颈部恶性肿瘤的40%，而5年生存率仅40%～50%[2]。组蛋白去乙酰化酶抑制剂（histone deacetylase inhibitors，HDACis）抑制（包括组蛋白）残留的赖氨酸蛋白的脱乙酰化，并调节各种基因的转录。近年来，已被临床用作抗癌药物和可能的抗糖尿病药物，但有证据表明高剂量的HDACis是有细胞毒性的[3-4]。针对组蛋白去乙酰化酶在癌症细胞中缺少调节这一弱点可作为治疗肿瘤细胞的新靶点[5]。本实验研究不同浓度的FK-228作用于口腔鳞癌Tca-8113后观察细胞增殖、凋亡，讨论组蛋白去乙酰化酶抑制剂对肿瘤细胞的作用。

1 材料与方法

1.1 材料

人舌癌鳞状细胞Tca-8113由上海交通大学赠予。FK-228采购自美国MCE公司，DMEM、胎牛血清采购自Gibco公司，PBS液由哈尔滨兽医研究所提供，CCK-8试剂盒由日本同仁采购，琼脂糖凝胶由哈尔滨兽医研究所提供，DNA Ladder凋亡试剂盒购自南京凯基生物公司。……
