pH示差法测定不同品系元宝枫叶片花青苷含量

摘 要:采集秋季转色后的元宝枫无性系叶片作为试验材料,采用pH示差法测定其不同品系叶片中花青苷的含量。结果显示:用恒温震荡培养箱25 ℃震荡提取6 h提取效果最好;pH示差法选定pH 值是1.0和4.5,提取液的平衡时间是60 min。试验的平均回收率是96.66%,由此可见,该方法能有效消除溶液中杂质对测定结果的影响,可用于元宝枫花青苷的定量分析。在所测量的5个品系中,花青苷含量最高的品系是1-7号,含量为0.845 3 mg·g-1,5个品系的平均花青苷含量是0.482 1 mg·g-1。

Abstract: In this study, the leaves of Acer truncatum Bunge were studied, and their total anthocyanin contents in different strains were investigated by pH-differential method. The results showed that using constant temperature oscillation incubator 25 ℃ oscillation extraction 6 hours it presented the best effect, and the pH selected were 1.0 and 4.0, and the buffering time was 60 minutes. By this method, the average recovery rate was 96.66%,therefore, this method can be used to the quantitative analysis of anthocyanins in Acer, and pH-differential method could eliminated the influence of impurities. Among the five strains,the content of anthocyanin in 1-7 was the highest ,which was 0.845 3 mg·g-1; the average anthocyanin content of these five strains was 0.482 1 mg·g-1.

元宝枫(Acer truncatum Bunge),槭树科(Aceraceae)槭属(Acer.L),是北方优良的园林绿化树种。秋季元宝枫叶色转为黄色、橙色或鲜红色,花青苷含量非常丰富[1]。本试验通过测定不同色系元宝枫叶片中花青苷含量,探讨叶色与花青苷含量的关系,为高含量花青苷品系的开发利用奠定理论基础。
花青苷广泛存在于大多数彩色植物体内,是水溶性类黄酮物质的一种,同时也是一种天然的食用色素,具有很多优点,如水溶性好、食用安全等,目前已普遍被消费者所接受。此外,在健康养生方面花青苷也起到不可忽视的作用:能缓解视觉疲劳、增强记忆力,同时还能清除体内过多的自由基[2],可以改善血液循环、预防心血管疾病、抗癌防癌[3]。元宝枫秋色叶中花青苷含量相当丰富,叶片中花青苷的开发利用价值潜力无限。
