高脂膳食联合链脲佐菌建立2型糖尿病大鼠模型研究

摘要：[目的]探索建立一种稳定简单的2型糖尿病大鼠模型。[方法]将40只无特定病原体级雄性大鼠采用随机数字表法随机分为造模组(30只)和对照组(10只)，造模组高脂饲料喂养后腹腔注射链脲佐菌(STZ)，对照组注射等体积缓冲液，监测两组空腹血糖、体重、饮水量、尿量等。[结果]造模组血糖和饮水量明显高于对照组(P<0.01)，成功造模25只，造模成功率83.33%。[结论]高脂膳食联合STZ腹腔注射诱导建立大鼠2型糖尿病模型操作简单、成功率较高，是一种较理想的造模方法。

Study on establishment of type 2 diabetic rat model using high fat diet combined with streptozotocin

AbstractObjective:To probe into a stable and simple model of type 2 diabetic rats.Methods:A total of 40 male rats without specific pathogen level were randomly divided into modeling group(30 rats) and control group (10 rats)by using a random number table method,the rats in modeling group were intraperitoneal injected with streptozotocin (STZ) after high-fat diet,while the rats in control group were injected with equal volume of buffer，monitoring the two groups of fasting blood glucose，weight，water intake，urine volume.Results:The blood glucose and water intake of the model group were significantly higher than that in control group(P<0.01)，the number of success establishment of model was 25，and its success rate was 83.33%.Conclusion:High fat diet combined with intraperitoneal injection of STZ to establish the model of type 2 diabetes rats was simple and had higher success rate,which was an ideal method of making model.

动物实验是生命科学的重要研究方法，其作为临床实验的过渡，从组织病理生理学的高度为临床护理技术的革新及安全应用提供了理论参考依据[1]。适当可行的动物实验非常必要，可有效指导护理理论与临床实践[2]。国际糖尿病联盟(IDF)统计显示，截止2014年全球共有3.87亿例糖尿病病人，其中2 型糖尿病(type 2 diabetes mellitus，T2DM)占85%～95%[3]。我国2010年调查数据显示，20岁以上人群中糖尿病总体患病率达到了9.7%，病人总数已超过9 200万例以上[4]。建立合适的动物模型是研究糖尿病发病机制和临床防治的基础。为了开展糖尿病足溃疡创面护理的动物实验研究，本研究采用高脂喂养后腹腔注射链脲佐菌(STZ)的方法建立了稳定的2型糖尿病大鼠模型。现报道如下。

1材料与方法

1.1实验动物及材料无特定病原体级雄性大鼠40只，由我校动物实训中心提供，体质量(200±15)g。高脂饲料(配方：蛋黄2.5%，蔗糖20.0%，猪油10.0%，基础饲料67.5%)喂养，自由摄食、进水，室温18 ℃～22 ℃。……
