PM2.5暴露对鸦胆子油乳治疗荷瘤肺癌小鼠腹腔巨噬细胞的作用

[摘要] 目的 探讨PM2.5暴露对鸦胆子油乳治疗荷瘤肺癌小鼠腹腔巨噬细胞的活性调节作用。 方法 建立肺癌荷瘤A549细胞小鼠模型32只,随机分为4组:治疗组1(鸦胆子油乳治疗组)和对照组1,治疗组2(PM2.5暴露联合鸦胆子油乳治疗组)和对照组2,每组8只。收集腹腔巨噬细胞,检测520 nm吸光度值。 结果 腹腔巨噬细胞吞噬能力显示,治疗组1高于对照组1,差异有统计学意义(P<0.05);治疗组2低于对照组2及治疗组1高于治疗组2,差异有统计学意义(P<0.01)。 结论 PM2.5暴露对鸦胆子油乳治疗荷瘤肺癌小鼠腹腔巨噬细胞的活性有抑制作用。

[Abstract] Objective To explore active regulating effect of PM2.5 exposure for peritoneal macrophages of tumor-bearing mice with lung cancer treated by bruceolic oil emulsion. Methods 32 mouse models in tumor-bearing A549 cell in lung cancer were randomly divided into 4 groups:treatment group 1 (bruceolic oil emulsion treatment group) and control group 1,treatment group 2 (PM2.5 exposure combined with bruceolic oil emulsion treatment group) and control group 2.Each group had eight mice.Peritoneal macrophages from these mice were collected and 520 nm absorbance degree value was detected. Results phagocytic ability of peritoneal macrophage indicated that treatment group 1 was higher than that of control group 1,and there was a statistical difference (P<0.05).The phagocytic ability in the treatment group 2 was lower than that of control group 2,and the phagocytic ability in the treatment group 1 was higher than that of treatment group 2, and there was a statistical difference (P<0.01). Conclusion PM2.5 exposure has inhibiting effect for activity of peritoneal macrophages in tumor-bearing mice with lung cancer treated by bruceolic oil emulsion.

空气污染物由多种资源消耗的途径产生,并可以损伤人体的健康和破坏自然环境。空气污染物由室内和室外污染物组成,包括基本颗粒(灰尘和碳颗粒),气体成分(一氧化碳、氮气、硫氧化物和臭氧),有机复合物,部分农药(二 [恶]辛类)和某些生物制品材料,当这些污染物进入呼吸系统时,就可能有潜在的毒性作用[1]。对人类的健康来说,颗粒混合物的成分是最主要的危险因素。相关的文献报道了颗粒物组分与日常死亡率和入院率的相关性[2-3]。针对空气颗粒物的研究不仅可以为某些疾病的预防工作奠定基础,也可以更好地阐述受到空气污染后相关疾病的致病机制,通过发现新的细胞反应,提高对细胞途径的理解,最终发展成新的有效的预防和治疗疾病的手段。

鸦胆子油乳,是由中药鸦胆子[Brucea javanica(L.)Merr.]干燥的成熟果实提取的脂肪油经乳化作用制成的水包油型乳剂,其主要活性成分油酸及亚油酸具有很强的抗癌活性,与肿瘤细胞膜有特异的亲和力[4]。研究表明,鸦胆子油乳可用于肺癌、食道癌、直肠癌、肝癌、前列腺癌等的治疗,对多种恶性肿瘤细胞均具有诱导凋亡的作用,可逆转多种癌细胞的多药耐药,临床上多采用鸦胆子油乳作为逆转剂与常用抗癌药合用治疗产生耐药性的肿瘤[5]。本研究主要探讨PM2.5暴露对鸦胆子油乳治疗荷瘤肺癌小鼠腹腔巨噬细胞活性调节的效果。

1 材料与方法

1.1 试剂和仪器

鸦胆子油乳:沈阳药大药业有限责任公司;脂多糖(LPS):Sigma公司;中性红:Amresco公司;0.4%台盼蓝染色液:北京索莱宝科技有限公司;A549细胞株购置于南京凯基生物发展有限公司;RPMI1640培养液:GIBCO公司;胎牛血清:浙江天杭生物科技股份有限公司;胰蛋白酶-EDTA消化液:GIBCO公司;青霉素链霉素溶液:Hyclone公司;磷酸缓冲液(PBS):Hyclone公司;75%乙醇;生理盐水。96孔板:康宁公司;100 mm细胞培养皿:康宁公司;15 ml离心管,康宁公司;50 ml离心管,康宁公司。
