莲子红皮多糖抗氧化与抑菌活性研究

摘要:以莲子(Semen nelumbinis)红皮磨皮粉为原料提取多糖,研究其抗氧化性及抑菌活性。研究发现莲子红皮多糖对羟自由基(·OH)的清除率达96.29%,对DPPH自由基(DPPH·)的清除率为41.79%,莲子红皮多糖的体外抗氧化活性低于维生素C;抑菌试验表明,莲子红皮多糖对大肠杆菌和金黄色葡萄球菌有明显的抑菌作用,最小抑菌浓度为20 mg/mL,在酸性条件下的抑菌作用更明显,且对大肠杆菌生长的抑制作用比金黄色葡萄球菌强,对黄曲霉和黑曲霉没有抑菌作用。

Abstract:The antioxidant and antimicrobial activities of the polysaccharides from red skin of locus seed were assessed in this paper. The study found that the polysaccharide on hydroxyl free radical (·OH) removal effect reached 96.29%, the highest DPPH· free radical (DPPH·) scavenging effect reached 41.79%. Through comparing with VC, the polysaccharide on free radical clearance rate was lower. The polysaccharide from red skin of locus seed had a strong antibacterial effect on the bacteria with the minimum inhibitory concentration of 20 mg/mL. Meanwhile,the antibacterial effect was stronger under acidic condition. The effect on colibacillus was more obvious than Staphylococcus aureus. And the fungus had no antibacterial effect on Aspergillus favus and Aspergillus niger.

莲子(Semen nelumbinis)是睡莲科植物莲的果实和种子。莲子是补脾止泻、益肾涩精、养心安神的常用中药,也是民间常用的滋补品。莲子含有丰富的多糖、磷脂、生物碱和类黄酮等营养保健成分,列入中国卫生部公布的首批既是食品又是药品的品种名单[1]。
莲子的加工一般要经过干燥、分级、去壳、除皮去心等工序,去皮一般采用手工除皮和机械磨皮两种方式。这两种去皮工艺生产率较低,损耗高。例如采用机械磨皮法,每加工100 kg红莲,就会磨去约15 kg红皮粉,目前这层红皮粉尚无有效的利用方式,由于味涩甚至不能作为牲畜饲料,只能大量堆于田间地头[2]。这些加工副产物莲皮粉含有多糖、黄酮、生物碱等多种生物活性成分,若能有效地开发利用,将变废为宝。
