头针结合艾灸对缺血缺氧性脑损伤幼鼠学习记忆能力及脑内BDNF的影响

摘要：目的观察头针结合艾灸对脑瘫幼鼠学习记忆能力的作用以及对脑源性神经生长因子(BDNF)的影响。方法7日龄清洁级新生SD幼鼠7窝共40只，随机分为假手术组(A组)、模型组(B组)、头针组(C组)、艾灸组(D组)和头针结合艾灸组(E组),造模成功后，采用头针、艾灸治疗2周后，对幼鼠的运动功能进行评价，行为学观察各组幼鼠Y迷宫实验结果;术后18 d通过免疫组化法检测大脑皮质BDNF阳性表达结果。结果假手术组Y迷宫实验耗时和BDNF阳性表达结果均低于模型组；各治疗组之间比较,Y迷宫实验结果头针结合艾灸组(67.71±7.60)优于头针组(74.73±8.06)(P=0.39<0.05)和艾灸组(81.18±10.1)(P=0.00<0.01);BDNF阳性表达结果头针结合艾灸组(20.46±2.03)与头针组(18.95±1.62)相比,P=0.43<0.05。结论治疗组均能改善幼鼠的学习记忆能力和提高BDNF阳性表达结果，以头针结合艾灸组效果最好；头针结合艾灸治疗可以提高BDNF水平的表达并发挥其生物活性，从而促进神经功能缺损的恢复，改善学习记忆能力。

Abstract:ObjectiveObserved scalp acupuncture combined with moxibustion on learning and memory ability of rats with cerebral palsy as wellas for brain-derived neurotrophic factor (BDNF) impact.Methodshypoxic-ischemic brain damage in young rats were randomly divided into sham operation group (A),model group (group B),scalp acupuncture group (group C),moxibustion group (group D) and Scalp combined with moxibustion group (E),Using scalp acupuncture,moxibustion treatment after two weeks,motor function of rats to evaluate the behavior observed in young rats in each group Y maze test results;18d after by immunohistochemical assay cerebral cortex BDNF expression results.ResultsThe sham group Y maze consuming and BDNF expression results were lower than the untreated group;Comparison,Y maze test results scalp acupuncture combined with moxibustion group(67.71±7.60) than among the treatment groups scalp acupuncture group (74.73±8.06)(P=0.39<0.05) and moxibustion group (81.18±10.1)(P=0.00<0.01),BDNF expression results scalp acupuncture combined with moxibustion group (20.46±2.03)compared with scalp acupuncture group (18.95±1.62),P=0.43<0.05.ConclusionThe treatment group rats can improve learning and memory ability and increase the BDNF expression results to scalp acupuncture combined with moxibustion group best effect;Scalp therapy combined with moxibustion can increase the level of expression of BDNF and exert their biological activity,thus contributing to the recovery of neurological deficit,improve learning and memory abilities.

新生儿缺氧缺血性脑损伤(hypoxic-ischemicbrain damage,HIBD)是由于出生前或出生时缺氧所造成的，患儿多留有脑瘫、癫痫等神经系统后遗症[1]。目前，对于新生儿缺氧缺血性脑损伤的有效治疗方法一直是国内新生儿科学的研究热点。本实验以脑源性神经生长因子(Brain-derived neurotrophic factor，BDNF)基因蛋白表达为指标，结合幼鼠学习记忆能力的观察，探讨头针结合艾灸治疗的脑保护作用机制。……
