超高效液相色谱—电喷雾串联质谱法检测茶叶中洛伐他汀的含量

摘要:采用超高效液相色谱-电喷雾串联四级杆质谱仪(UPLC-ESI-MS/MS),于多反应监测(MRM)模式下建立了茶叶中洛伐他汀的定性定量分析方法。样品经甲醇超声提取,Waters Acquity UPLC?誖BEH C18(1.7 μm,2.1 mm×50 mm)柱分离,超高效液相色谱-串联质谱法测定。在0.000 8~0.200 0 μg/mL间有良好的线性关系(R2>0.999 0),检出限为0.000 2 μg/mL;在3个添加浓度水平下,平均加标回收率为98.54%~ 109.86%;相对标准偏差(RSD,n=6)为1.45%~3.09%。该方法准确、可靠、灵敏度高,适用于茶叶中洛伐他汀含量的检测。

Determination of Lovastatin in Tea Using Ultra Performance Liquid Chromatography-Electrospray Tandem Mass Spectrometry

Abstract: An UPLC-ESI-MS/MS method with multiple reaction monitoring (MRM) mode was developed for the qualitative and quantitative analysis of lovastatin in tea. The sample preparation was based on ultrasonic-assisted extraction,Waters Acquity UPLC?誖BEH C18(1.7 μm,2.1 mm×50 mm) column separation and UPLC-MS/MS determination. The method showed a good linearity (R2>0.999 0) in the 0.000 8~0.200 0 μg/mL and LOD of 0.000 2 μg/mL. For three spiked levels,the average recoveries were from 98.54% to 109.86%, and the relative standard deviations (RSD,n=6) were from 1.45% to 3.09%. The method was reliable, accurate, sensitive and appropriate for the determination of lovastatin in tea.

1979年, Endo等[1]从Monascus ruber的培养液中分离出抑制HMG-CoA(3-羟基-3-甲基戊二酸单酰辅酶A)还原酶活性的Monacolin K,同时 Alberts等[2]于1980 年也从Aspergillus terreus中分离出与Monacolin K相同的物质Mevinolin,后命名为洛伐他汀(Lovastatin),由Merk公司率先制成药品上市。洛伐他汀是人体胆固醇合成中的关键酶HMG-CoA还原酶的抑制剂,因其疗效好、副作用低,目前已被广泛地应用于临床高血脂症等疾病的治疗。
普洱茶是以云南大叶茶[Camellia sinensis (Linn.) var. assamica (Masters) Kitamura]的晒青毛茶为原料,在微生物的作用下,经漫长的自然陈化或快速人工渥堆后发酵而成。目前,能产洛伐他汀的微生物已见报道[3],如曲霉菌(Aspergillus)、红曲霉菌(Monascus)、毛霉菌(Mucor)和青霉菌(Penicillium)等。Hwang等[4]首先在普洱茶中发现有洛伐他汀的存在;随后,Yang等[5]研究指出洛伐他汀是在普洱茶中发现的惟一的他汀类化合物,在乙酸乙酯提取物中含量较高,且全部以内酯环结构存在,而在100 ℃普洱茶水提取物中含量较低,以内酯环结构和羟酸结构两种形式存在;谢春……
