不同产地及炮制方法雪上一枝蒿多糖含量分析

摘要:目的 对雪上一枝蒿中多糖的含量测定方法进行研究,建立紫外分光光度法测定雪上一枝蒿多糖的含量测定方法,并考察不同产地、不同炮制方式对多糖含量的影响。方法 用80%乙醇回流去除还原性糖等杂质,后热水提取多糖,采用苯酚-硫酸比色法于488 nm波长处用紫外-分光光度法测得吸光度值,从而得出其多糖的含量。结果 雪上一枝蒿多糖以无水葡萄糖计,在0.003~0.015 mg/mL范围内呈良好的线性关系(R=0.9999),回收率为99.35%~99.85%(n=6),RSD值为0.18%。结论 本法准确、可靠,重现性好,适用于测定雪上一枝蒿中多糖含量;不同产地不同批次药材中的多糖含量存在一定差异;炮制方式对雪上一枝蒿多糖含量无显著影响。

【Abstract】Objective: To observe the expression of HSP60 and Caspase-3 of acupuncture in rats with cerebral infarction and to preliminarily explore the protective mechanism of acupuncture on neuronal apoptosis in rats with ischemic stroke. Methods: The cerebral ischemia rats were induced by middle cerebral artery suture to make reperfusion injury models and randomly divided into sham operation group, model group, electroacupuncture point group, electroacupuncture non-meridian non-acupoint group and moxibustion acupoint group. After the intervention of electroacupuncture or moxibustion, the neurological deficit score and postural reflex score of the rats were evaluated at 0 and 12 hours after the operation. The expression levels of HSP60 and Caspase-3 mRNA were detected by fluorescence quantitative PCR. Results: The nerve function of the rats in the electro-acupuncture acupoint group and moxibustion acupoint group was better than that of the model group, and the difference was statistically significant(P<0.05). Compared with that of the model group, the HSP60 mRNA of the electro-acupuncture acupoint group increased significantly, and the difference was significant(P<0.05) while the expression of Caspase-3 mRNA did not change significantly. Conclusion: Electroacupuncture can improve the neurological function of rats with cerebral ischemia-reperfusion injury. The mechanism may be related to the up-regulation of the expression of HSP60 in brain tissue and mediating anti-neuronal cell apoptosis.

雪上一枝蒿為毛茛科植物短柄乌头Aconitum brachypodum Diels.的干燥块根[1]。其味苦、辛,性温,有大毒,主产于云南北部的东川、会泽等地,具有祛风镇痛的功效,用于治疗风湿疼痛、跌打损伤等症。雪上一枝蒿含有乌头碱、次乌头碱、雪上一枝蒿甲素等多种生物碱和β-谷甾醇、胡萝卜苷等非生物碱成分,其中生物碱主要是二萜生物碱,目前认为是其主要成分,雪上一枝蒿具有镇痛、局部麻醉、抗炎和抗肿瘤等药理作用[2]。以雪上一枝蒿为原料,开发了包括雪上一枝蒿注射剂、骨痛灵酊、云南白药气雾剂、云南白药膏、肿痛气雾剂等多种制剂,是云药产业发展中一个很具特色的品种。……
