IRF5通过对巨噬细胞极化的调节影响脂质摄取及排出的探究

【摘要】动脉粥样硬化(AS)是严重危害人类生命健康的一類慢性血管疾病,局部胆固醇的大量蓄积和炎症的发生是其主要原因。现已证明许多细胞因子通过调节巨噬细胞向M1及M2型的转化,影响巨噬细胞对脂质的摄取及排出。该文将阐述干扰素调节因子5(IRF5)是否通过调节巨噬细胞的极化而对巨噬细胞的脂质摄取及排出产生影响,从而为以IRF5为作用靶点治疗AS的可能性提供一些前期理论研究。

【Abstract】Atherosclerosis is a chronic vascular disease that seriously endangers the life and health of human. The accumulation of local cholesterol and the recurrence of inflammation are the major causes.Through the transformation to M1 and M2, macrophages play an important role in regulating the uptake and excretion of lipid.In this article, whether the interferon regulatory factor 5 (IRF5) can influence the function of uptake and excretion of lipid of the macrophage by regulating the polarization of macrophage or not was discussed,aiming to explore the possibility of IRF5 as a target for the treatment of atherosclerosis.

由动脉粥样硬化(AS)引起的急性心脑血管事件的发病率和致死率呈逐年上升的趋势,其发病基础以大量蓄积的脂质和难以控制的炎症为主。巨噬细胞在调节脂质及炎症反应中发挥着重要作用。本文主要阐述巨噬细胞摄取及排出脂质的过程及干扰素调节因子5(IRF5)是否通过对巨噬细胞极化的调控进一步影响此过程,探讨治疗AS的新靶点。
一、IRF5可能通过干扰巨噬细胞内脂质的摄取及排出影响AS
动脉内皮下沉积的脂质吸引单核细胞迁移、附着并逐渐分化成巨噬细胞,随后其细胞表面的清道夫受体(SR)摄取氧化型低密度脂蛋白(oxLDL)进入细胞内,经胆固醇酰基转移酶-1和中性胆固醇酯水解酶的酯化、水解等处理后转化为游离胆固醇,堆积在巨噬细胞内。正常情况下,巨噬细胞内约80%的游离胆固醇经膜孔扩散流出细胞外,还有一部分游离胆固醇在细胞中被胆固醇转运蛋白转移至细胞外。……
