【摘 要】 目的：观察金蛭方对缺血性中风大鼠血清及脑组织超氧化物歧化酶（SOD）活性和丙二醛（MDA）含量的影响，探讨其治疗缺血性中风的作用机制。方法：采用线栓法复制缺血性中风大鼠模型，随机分为假手术组、模型对照组、金蛭方高剂量组、金蛭方低剂量组、阳性药尼莫地平片对照组。测定大鼠血清和脑组织SOD活性和MDA含量。结果：模型组大鼠血清、脑组织中SOD活性较假手术组明显降低（P<0.01）；MDA含量均较假手术组明显升高（P<0.01）。用药后各治疗组大鼠血清和脑组织中SOD活性明显升高（P<0.05，P<0.01）；MDA含量明显降低（P<0.05，P<0.01）。高、低剂量组在增强SOD活性和降低MDA含量方面的作用优于对照组（P<0.05）。结论：金蛭方通过调节缺血性中风大鼠SOD活性、MDA的含量以达到抗缺血性中风作用。

Abstract： ObjectiveObsevve the Jinzhi Fang for ischemic stroke in rats serum and brain tissue superoxide dismutase （SOD） activity and malondialdehyde （MDA） content， the influence of its action mechanism for the treatment of ischemic stroke. Methods By using line switch copy of ischemic stroke in rats model， were randomly divided into control group， model control group， the Jinzhi Fang high dose group， gold Jinzhi Fang low dose group， positive medicine nim flat piece of the control group. Determination of rats serum and brain tissue SOD activity and MDA content. Results The model group rats serum and brain tissue SOD activity significantly decreased in the control group （P<0.01）； MDA content were increased significantly in the control group （P<0.01）. After each treatment group rats serum and brain tissue SOD activity significantly increased （P<0.05， P<0.01）； The MDA content significantly decreased （P<0.05，P<0.01）. High and low dose group was increase SOD activity and reduce MDA content of the role of is better than that of control group （P<0.05）. Conclusion Jinzhi Fang by adjusting the ischemic stroke SOD activity and MDA content of rats in order to achieve the function of ischemic stroke.

缺血性中风约占中风病的30%，其发病率高、致残率高和死亡率高，给家庭及社会造成极大负担。氧化应激学是缺血性脑血管病的病理生理机制之一[1]。金蛭方系课题组经过10余年临床筛选出的治疗缺血性中风的效方，拟通过建立大鼠急性脑缺血再灌注模型，观察金蛭方对大鼠血清及脑组织中SOD活性及MDA含量的影响，以探讨其抗氧自由基水平方面的可能作用机制。

1 材料与方法

1.1 动物 健康雄性SD大鼠60只，清洁级，体重250～300 g，由河北省实验动物中心提供，合格证号：1119069。

1.2 药物与试剂 金蛭方组成：水蛭、郁金、川芎、冰片（由迁安市中医医院制剂室制备），上4味除冰片外水提浓缩成高、低剂量后加入相应研磨冰片，所含生药分别为：0.30 g/mL（高剂量）、0.15 g/mL（低剂量）。尼莫地平片（国药准字：H41022175，规格：20 mg/片，郑州瑞康制药有限公司）配制成0.001 g/mL水溶液。

水合氯醛注射液（批号：20120206，上海化学试剂公司）；超氧化物歧化酶（SOD）試剂盒（批号：20130704）、丙二醛（MDA）试剂盒（批号：20130709）均购自南京建成生物工程研究所。……
