[摘要] 目的 应用两种荧光原位杂交（FISH）试剂盒检测羊水染色体，比较两者在临床的应用效果。 方法 采用北京金菩嘉和美国雅培Vysis生产的FISH试剂盒对30例未培养的羊水进行染色体检测，比较两者在信号强度和杂交率的差异。 结果 30例样本均获得FISH检测结果，且两种试剂临床诊断符合率达到100%，杂交率无统计学意义（P>0.05）。Vysis试剂21/13染色体荧光杂交信号较强，金菩嘉试剂18染色体信号清楚，不易融合。 结论 Vysis与金菩嘉两种FISH检测试剂应用染色体异常产前诊断均可达到同样的临床诊断效果。

Comparative study on application of two fluorescence in situ hybridization （FISH） kits for inspecting amniotic fluid chromosomes

[Abstract] Objective Two fluorescence in situ hybridization （FISH） kits were utilized to inspectamniotic fluid chromosomes，to compare the both kits in clinical application. Methods Two fluorescence in situ hybridization （FISH） kits from Beijing GPmedical and Abbott were utilized to inspect 30 samples of amniotic fluid chromosomes.Their differences in respect of signal intensityand hybridizing were compared. Results All samples of the 30 cases were successfully detected by the both FISH kits.The hybridizing ratethere was no significant difference （P>0.05）.Vysis kits 21/13 chromosome fluorescent hybridization signal was stronger than the GP kits.The GP kits 18 chromosomes signal was clear and not easy to fuse. Conclusion the FISH kits from the Beijing GPmedical and Abbott Vysis can make the same prenatal diagnosis for the chromosome abnormality.

熒光原位原位杂交技术（FISH）基于分子细胞遗传学发展的学科，始于1998年，至今在临床研究中广泛应用，在产前诊断领域更是不断完善和推广[1-3]。由于商业试剂盒应用方便、高效、稳定且专业化，临床上更青睐采用FISH商业试剂盒进行产前诊断[4]，如美国雅培Vysis公司（美国），其试剂以极高的灵敏度及可重复性在临床应用中极受欢迎。目前我国金菩嘉公司（中国）自行研发成功FISH商业试剂盒可满足产前诊断系列使用。本课题通过雅培Vysis公司和金菩嘉公司分别研制的FISH试剂盒进行临床应用比较，以便国产FISH商业试剂盒的优化，增强市场竞争力。

1 资料与方法

1.1 一般资料

随机选取2009年3～10月在南方医院行产前诊断的高危孕妇30例，年龄18～44岁，平均（30.4±5.3）岁。孕周17～37周，平均（23.1±4.4）周。产前诊断指征包括：（1）孕妇年龄≥35岁10例；（2）母血清唐氏筛查高风险15例；（……
