胞核内高表达M—CSF对HeLa细胞生长增殖的影响

[摘要] 目的 分析人宫颈癌HeLa细胞核内M-CSF增高对化疗药物的影响。 方法 体外培养HeLa细胞、HeLa-M细胞以及转染M-CSF空载体HeLa-C细胞。分别使用紫杉醇和顺铂(浓度分别为0.3、3、30 μg/mL)作用48 h,采用MTT方法比较细胞的相对活性,Westen blot分析转染M-CSF对抗凋亡分子Bcl-2表达的影响。 结果 当两种化疗药物的浓度为3、30 μg/mL时,与HeLa-C细胞和HeLa细胞相比,HeLa-M细胞活性明显增高,差异有统计学意义(P<0.05);当浓度为0.3 μg/mL时三种细胞活性无明显差异。Westen blot结果显示,转染M-CSF后能够使细胞中的抗凋亡蛋白Bcl-2表达水平明显升高。 结论 人宫颈癌HeLa细胞核中的M-CSF增高可以使细胞的药抗性提高,从而抑制HeLa细胞的凋亡。

[Abstract] Objective To analyze the effect of elevated M-CSF in nuclei on the chemotherapeutic drugs of human cervical cancer HeLa cells. Methods HeLa cells, HeLa-M cells and HeLa-C transfected with M-CSF empty vector were cultured in vitro. The paclitaxel and cisplatin(concentrations were 0.3, 3, 30 μg/mL) were used for 48 h, respectively. The relative viability of the cells was compared by MTT method. The effect of transfection with M-CSF on the expression of Bcl-2 was detected by Western blot analysis. The effect of paclitaxel and cisplatin (concentration 0.3, 3, 30 μg/mL) on the expression of anti-apoptotic molecule Bcl-2 was detected by MTT method. Results The viability of HeLa-M cells was significantly higher than that of HeLa-C cells and HeLa cells when the concentration of the two chemotherapeutic drugs was 3, 30 μg/mL, and the difference was statistically significant(P<0.05). There was no significant difference in cell viability between the three kinds of cells when the concentration was 0.3 μg/mL. The results of Western blot showed that the expression of anti-apoptotic protein Bcl-2 was significantly increased after transfection with M-CSF. Conclusion The increase of M-CSF in the nucleus of human cervical cancer HeLa cells can improve the drug resistance of the cells and inhibit the apoptosis of HeLa cells.

M-CSF即巨噬細胞集落刺激因子,简称集落刺激因子[1],主要是由链间二硫键所形成的同源二聚体糖蛋白,其与CSF-1R相互作用会使受体发生二聚化反应,随后可以与细胞内的多条信号通路发生相应的作用,比如JAK/STAT等,与不同细胞作用会产生不同的生物学效应[2-3]。2006年时相关学者以小鼠为例,使用抗M-CSF逆转了种植在小鼠身上的MCF-7细胞,这说明在治疗乳腺癌中,抗M-CSF是一种潜在的治疗方法[4]。对妇女而言,宫颈癌是第二种常见的恶性肿瘤疾病,临床上的常见药物为顺铂类药物,但是容易产生耐药性[5-6]。因此,对人宫颈癌HeLa细胞核内的M-CSF研究具有深远的意义。本研究对其进行相应研究,现报道如下。
1 资料与方法
1.1 一般资料
从北京医科大学细胞库中购入人宫颈癌HeLa细胞,转染的HeLa-C细胞以及转染M-CSF细胞等需要在前期通过实验转染得到。所使用的MTT从Sigama公司购买。顺铂从哈尔滨三联制药公司购入,紫杉醇为齐鲁制药公司生产。DMSO由美国Ameresco公司生产,低温高速离心机从德国引入,生化培养箱为SANYO公司生产,其他试剂为我国国内生产。……
