LDH法测定纤维蛋白胶对人脂肪干细胞的细胞毒性

[摘要] 目的 采用LDH法测定纤维蛋白胶对人脂肪干细胞的细胞毒性。 方法 取成人吸脂术后的废弃脂肪组织并从中得到人脂肪干细胞,分别制成50%浸提液组、100%浸提液组、阴性对照组、阳性对照组,并采用LDH检测试剂盒测定各组纤维蛋白培养液中LDH的含量。 结果 原代ASCs生長缓慢,传代后生长加速,且ASCs呈成纤维样细胞形态;24 h和72 h两个时间点中50%纤维蛋白胶浸提液组、100%纤维蛋白胶浸提液组分别与阴性对照组比较,差异无统计学意义(P>0.05)。 结论 纤维蛋白胶对ASCs无明显细胞毒性,可为以纤维蛋白胶为支架材料的脂肪组织工程研究提供参考。

[Abstract] Objective To determine the cytotoxicity of fibrin glue to human adipose stem cells by LDH method. Methods The abandoned adipose tissue of human liposuction was taken and the human adipose stem cells were obtained and made into the 50% leaching solution group, 100% leaching solution group, negative control group and positive control group.And the LDH test kit was used to detect the content of LDH in each fibrin culture medium. Results The growth of primary ASCs was slow. The growth of ASCs was accelerated after passage, and ASCs appeared fibroblast-like morphology. There was no significant difference between 50% fibrin glue extract group ,100% fibrin glue extract group and the negative control group at 24 and 72 hours(P>0.05). Conclusion Fibrin glue has no obvious cytotoxicity on ASCs, and it can give a reference to the study of adipose tissue engineering with fibrin glue as scaffold.

软组织缺损是整形外科临床工作中经常遇到的问题,中老年人面部软组织容量减少的矫正也有着广泛需求。目前临床上常用的作为软组织填充的材料可大致分为两类:一类是暂时性填充材料,它们一般是人工合成的机体组织中的正常成分,如透明质酸或胶原蛋白,其因具有生物相容性好、可被降解吸收的优点而应用广泛,但也存在填充效果维持时间短的缺点;另一类是永久性填充材料,它们一般是人工合成的惰性材料组成,如硅胶,其作为软组织填充具有异物植入、效果不自然的缺点[1,2]。自体脂肪颗粒移植近年来发展较快,但移植后的脂肪颗粒的存活尚不理想,导致移植存活率较低,需要多次手术才能达到需要的填充效果[3,4]。近……
