高效液相色谱法测定白驳丸中4种重楼皂苷含量

目的 建立高效液相色谱法测定白驳丸中4种重楼皂苷的含量。方法 采用Agilent Zorbax SB C18色谱柱(250 mm×4.6 mm，5 μm)；柱温35 ℃；流动相A为乙腈，流动相B为水，进行梯度洗脱；流速1.0 mL·min-1，检测波长203 nm。结果 4种重楼皂苷质量浓度在18.12～465.9 μg·mL-1范围内呈良好线性关系(r=0.999 9)；平均回收率在95.11%～99.74%之间；RSD值均小于3.0%。结论 该方法灵敏度高、准确性好、具有较好的重复性和稳定性，可用于白驳丸的质量控制。

A.stract:Objective To establish an HPLC method for simultaneously determining the contents of 4 polyphyllins in Baibo Pills. Methods HPLC system consisted of Agilent Zorbax SB C18Column， gradient eluted with acetonitrile-water solution， the flow rate of the mobile phase was 1.0 mL·min-1， UV detection wavelength was set at 203 nm and column temperature was maintained at 35 ℃.Results 4 polyphyllins showed good linearity within the tested concentration scope(18.12-465.9 μg·mL-1，r=0.999 9)；the method was validated and the recoveries of 4 polyphyllins were 95.11%～99.74%；RSD<3.0%. Conclusion This method is simple， accurate and reproducible，and can be used for the quality control for Baibo Pills.

白驳丸为徐州市中医院皮肤科常用制剂，具有补肾健脾、养血通络、祛风退斑的作用；已在临床使用多年，对白癜风、脂溢性脱发有显著的疗效。白驳丸组方药味较多，主要由重楼等中草药复方组成， 成分比较复杂。重楼为百合科植物云南重楼ParispolyphyllaSmith var．yunnanensis(Franch.) Hand.-Mazz. 或七叶一枝花ParispolyphyllaSmith var．chinensis(Franch.)Hara 的干燥根茎[1-2]；味苦，微寒，有小毒；归肝经；清热解毒，消肿止痛，凉肝定惊，主要用于疗疮痈肿[3]，是一种临床常用的皮肤科用药。重楼主含甾体皂苷类成分，约占化合物总含量的80%[4]。

为有效监测该制剂的产品质量，本研究采用高效液相色谱法测定重楼中4种皂苷含量，所得定性、定量分析方法可客观地反映本制剂的内在品质，为白驳丸的质量控制和评价指标提供了理论依据[5-7]。

1 仪器与试药

1.1 仪器 高效液相色谱仪(Waters e2695-2998，PDA检测器，Empower 3色谱工作站)；……
