响应面法对栗蘑多糖液体发酵培养基的优化

摘 要:该文采用统计学方法对栗蘑虹灰1号菌株进行液体发酵配方优化,以得到适合栗蘑多糖发酵的最佳培养基配比。以栗蘑多糖含量为指标,首先采用前期单因素实验得出的6个因素,再进行Plackett-Bumrman设计筛选出影响栗蘑多糖含量的关键因素,通过响应面法确定关键因素的最佳用量和配比。结果表明:栗蘑菌株虹灰1号液体发酵培养基的最佳配方为:黄豆粉15g/L、玉米粉17.25g/L、葡萄糖20g/L、磷酸二氢钾2.237g/L、硫酸镁2g/L、蛋白胨5.88g/L,并在pH自然、25℃、140r/min条件下培养,比优化前产量提高了76.03%。

Abstract:The objective of the present study was the optimization of liquid fermentation medium for producing polysaccharide by Chestnut mushroom by the single factor tests and response surface methodology. Overall research was preceded by rational design. Technological condition parameters were determined according to regression analysis by Design-Expert 7.1.6. The RSM is feasible for optimum extraction process and this will improve the liquid fermentation medium for producing polysaccharide . The optimum condition was as follows:soybean powder 15g/L,corn flour 17.25g/L,glucose 20g/L ,KH2PO4 2.237g/L,MgSO4 2g/L and peptone 5.88g/L,pH7.0,25℃,140r/min,production increased by 76.03% than before optimization.

栗蘑,又名灰树花、舞茸,是一种集食用和药用于一体的菇类,味道和口感俱佳,并且具有良好的药效,其中栗蘑多糖具有很好的抗癌作用[1]。栗蘑多糖是由葡聚糖及少量蛋白质的异聚糖复合物组成的,主要是以通过β-(1-6)糖苷键结合为主链的葡聚糖和以通过β-(1-3)糖苷键结合为主链的葡聚糖的混合活性葡聚糖。传统提取栗蘑多糖的方法是从野生栗蘑中获得,然而野生栗蘑数量有限,并且只有在达到一定树龄的老树根部才会生长,难以规模化生产[2-3]。而人工栽培栗蘑子实体同样存在生产周期长、产量有限的问题。通过现代的液体发酵培养技术可以在短时间内获到大量的栗蘑菌种的发酵产物,研究表明,液体发酵培养得到的栗蘑菌丝体与野生或人工栽培得到的栗蘑在化学成分或生理功能上均很相似,并且发酵液中含有较多的栗蘑多糖。……
