过表达糖原合成酶1对非小细胞肺癌细胞增殖能力的影响

【摘要】目的 探讨糖原合成酶(GYS1)对非小细胞肺癌细胞增殖能力的影响。方法 采用实时荧光定量PCR(qRT-PCR)检测正常人支气管上皮样细胞系HBE细胞和非小细胞肺癌细胞系NCI-H661、NCI-H1299中GYS1 mRNA相对表达量。过表达NCI-H1299细胞的GYS1,构建GYS1过表达细胞模型,采用MTT增殖实验、EdU增殖实验评估细胞的增殖能力,应用平板克隆形成实验评估细胞的克隆形成能力。结果 人非小细胞肺癌细胞NCI-H661、NCI-H1299中GYS1 mRNA的表达水平高于人正常支气管上皮样细胞HBE的表达水平(P均< 0.001)。NCI-H1299-GYS1过表达细胞的GYS1 mRNA和蛋白相对表达量均上调(P均< 0.001)。过表达GYS1促进了NCI-H1299细胞的增殖和克隆形成(P均< 0.001)。结论 GYS1在非小细胞肺癌中高表达,过表达GYS1促进了非小细胞肺癌细胞的增殖。

【Abstract】Objective To evaluate the effect of glycogen synthase 1 (GYS1) on the proliferation of non-small cell lung cancer cells. Methods The relative expression levels of GYS1 mRNA in normal human bronchial epithelial cell line HBE cells, non-small cell lung cancer cell lines NCI-H661 and NCI-H1299 were quantitatively detected by quantitative real-time fluorescence PCR (qRT-PCR). The NCI-H1299 cells overexpressing GYS1 were utilized to establish the GYS1 overexpression cell model. MTT assay and EdU proliferation experiment were adopted to evaluate the cell proliferation ability, and plate clone formation assay was employed to assess the cell clone formation ability. Results The expression levels of GYS1 mRNA in human non-small cell lung cancer cells NCI-H661 and NCI-H1299 were significantly higher than that in normal human bronchial epithelial cell HBE (both P < 0.001). The relative expression levels of GYS1 mRNA and protein were significantly up-regulated in the NCI-H1299-GYS1 overexpressing cells (both P < 0.001). Overexpression of GYS1 remarkably promoted the proliferation and clone formation of NCI-H1299 cells (both P < 0.001). Conclusions GYS1 is highly expressed in non-small cell lung cancer. Overexpression of GYS1 promotes the proliferation of non-small cell lung cancer cells.

肺癌是人類健康的主要威胁之一,据估计2018年全球肺癌死亡人数达到180万[1]。肺癌的病理学类型常见非小细胞肺癌和小细胞肺癌。85%的肺癌为非小细胞肺癌,大多数患者在发现时已处于中晚期,失去了手术机会[2]。非小细胞肺癌发生发展关键驱动基因的深入研究,对患者和临床医师而言具有重要的意义。

代谢紊乱是许多肿瘤的特征。肿瘤对其代谢产物进行重新编程,以产生足够的能量维持生存。糖原主要储存在肌肉和肝脏中,在需要时作为能量供应。然而,非小细胞肺癌糖原代谢的改变及其分子机制仍然有待于进一步研究。糖原合成的关键酶之一糖原合成酶(GYS)包括2种亚型,即GYS1和GYS2。GYS1通常在肌肉、肺、心脏和肾脏中表达,而GYS2主要局限于肝脏[3]。研究表明,GYS1在缺氧条件下被迅速诱导,并与胶质母细胞瘤、乳腺癌和结肠癌细胞系中的糖原积累呈正相关[4]。GYS1的异常表达不仅降低了细胞糖酵解,而且增加了糖原反应性AMP激酶的激活,导致髓系白血病细胞的生长抑制[5]。……
