双藤痹痛凝胶膏剂对胶原诱导性关节炎模型大鼠血浆炎性因子和滑膜组织相关因子的影响

摘要:目的 观察双藤痹痛凝胶膏剂对胶原诱导性关节炎(CIA)模型大鼠血浆炎性细胞因子和滑膜组织Toll样受体4(TLR4)、髓样分化因子88(MyD88)、核因子-κB(NF-κB)表达的影响,探讨其相关作用机制。方法 大鼠尾部皮下注射胶原乳剂制作CIA模型。大鼠随机分为正常组、模型组和双藤痹痛凝胶高、低剂量组,ELISA检测血浆肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β及IL-6的含量,RT-PCR检测滑膜组织TLR4、MyD88、NF-κB的mRNA表达,Western blot检测滑膜组织TLR4、MyD88、NF-κB的蛋白表达。结果 与模型组比较,双藤痹痛凝胶高、低剂量组大鼠血浆TNF-α、IL-1β及IL-6含量显著降低,滑膜组织TLR4、MyD88、NF-κB mRNA和蛋白表达明显降低。结论 双藤痹痛凝胶膏剂可能是通过抑制炎症反应、调节滑膜组织TLR4/NF-κB信号通路相关靶点表达发挥抗炎作用。

Effect of Shuangteng Bitong Cataplasm on Inflammatory Cytokines in Plasma and Relevant Factors of Rats with Collagen-induced Arthritis

Abstract: Objective To study the effect of Shuangteng Bitong Cataplasm on TLR4, MyD88 and NF-κB and inflammatory cytokines in rats with collagen-induced arthritis; To discuss relevant mechanism of action. Methods A CIA model was made by subcutaneous injection of collagen emulsion from rat tail. Rats were randomly divided into normal group, model group, Shuangteng Bitong Cataplasm high-dose and low-dose groups. The contents of TNF-α, IL-1β and IL-6 in plasma were evaluated by ELISA, and the expression levels of TLR4, MyD88 and NF-κB in synovial tissues were evaluated by RT-PCR and Western blot. Results Compared with the model group, the contents of TNF-α, IL-1β and IL-6 in serum decreased remarkably in the Shuangteng Bitong Cataplasm low-dose and high-dose group, and the expression levels of TLR4, MyD88 and NF-κB in synovial tissues decreased remarkably. Conclusion Shuangteng Bitong Cataplasm can inhibit inflammatory reaction and regulate the expressions of relevant factors in synovial tissues to realize the anti-flammatory effects.
