金莲花总黄酮对人甲状腺乳头癌K1细胞三叶因子3表达变化的影响

【摘要】目的探讨金莲花总黄酮对人甲状腺乳头癌K1细胞增殖抑制作用及三叶因子3(TFF3)表达变化。方法不同浓度金莲花总黄酮作用于体外培养K1细胞24、48、72 h，MTT检测细胞生长抑制率，Western blotting和Real-time PCR检测不同浓度总黄酮对K1细胞TFF3蛋白及mRNA表达的影响。结果MTT实验结果表明，同一时间总黄酮对K1细胞生长增殖抑制率随着药物浓度的增高而升高(P＜0.05)，同一浓度总黄酮对K1细胞抑制率随时间延长而升高(P＜0.05)。Western blot及QPCR结果显示随着金莲花总黄酮浓度的升高K1细胞TFF3蛋白及mRNA水平下降(P＜0.05)。结论金莲花总黄酮对人甲状腺乳头癌细胞具有抑制作用，并能抑制TFF3的表达。

【ABSTRACT】Objective To investigate the inhibition effect of Trollius flavonoids on the proliferation of Trefoil factor 3(TFF3)in human papillary thyroid cancer K1 cells in vitro，and to explore its effect on the expression of TFF3 in K1 cells.Methods The K1 cells cultured in vitro were treated by different concentrations of Trollius flavonoids for 24h，48h，72h，respectively，then the inhibition rate on K1 cells growth was dectected bymTT.The changes of expression of TFF3 protein/mRNA were detected by Western blotting and Real-time PCR，respectively.ResultsmTT detection showed that Trollius flavonoids had inhibition effects on proliferation and growth of K1 cells in a dose and time dependentmanner(P＜0.05).The examination results by Western blotting and Real-time PCR showed that the expression levels of TFF3 protein andmRNA of K1 cells were decreased with the increase of flavonoids concentration(P＜0.05).Conclusion Trollius flavonoids have the inhibition effect on human throid papillary cancer cells and can inhibit the expression of TFF3.
