Oct4和Sox2在食管鳞癌组织中的表达及相关性探讨

摘要：目的探讨Oct4和Sox2在食管鳞癌组织中的表达及二者之间的相关性。方法应用免疫组化SP法和流式细胞术检测98例食管鳞癌标本及43例正常食管黏膜组织中Oct4和Sox2的表达情况，分析其与各临床病理特征的关系及二者之间的相关性。结果食管鳞癌组织Oct4和Sox2的阳性表达率均高于正常组织(P<0.05)；流式细胞术结果显示，癌组织中Oct4和Sox2的蛋白相对含量也明显高于正常组织(P<0.05)。随着分化程度的降低，Oct4和Sox2的表达明显升高，即二者与食管癌组织的分化程度呈负相关(P<0.05)，且Oct4和Sox2的表达与TNM分期、有无外膜浸润明显相关(P<0.05)，而与患者年龄、性别无关。食管鳞癌组织中Oct4和Sox2表达呈正相关(r=0.295，P=0.003)。结论Oct4和Sox2在食管鳞癌的发生、发展过程中起重要作用，有望成为食管癌生物治疗的新靶点。

Expressions of Oct4 and Sox2 Protein and Their Correlations

in Esophageal Squamous Cell Carcinoma

Abstract:ObjectiveTo study the expression of Oct4 and Sox2 proteins and their correlations in esophageal squamous cell carcinoma(ESCC).MethodsImmunohistochemistry staining SP and folw cytometry(FCM)were used to detect the expression of Oct4 and Sox2 in 98 cases of ESCC and 43 normal mucous memebranes.Subsequently the relationship between postoperative pathological features was analyzed with the expression of Oct4 and Sox2.ResultsThe expressions of Oct4 and Sox2 in ESCC were obviously higher than that in normal esophageal(P<0.05).For the protein relative level detected by FCM,there was a significant deference between the ESCC and normal esophageal of the protein content of Oct4 and Sox2(P<0.05).With the loss of differentiation degree,the expressions of Oct4 and Sox2 obviously rose in ESCC.The correlation is negative between Oct4 and Sox2 with the differentiation degree(P<0.05).The expressions of Oct4 and Sox2 were positively correlated to TNM stage and invasion of adventitia(P<0.05).Oct4 expression in ESCC was positively correlated with that of Sox2(r=0.295，P=0.003).ConclusionOct4 and Sox2 play crucial roles in occurrence and progression and are useful for the target therapy of ESCC.

Oct4(octamer-binding transcription factor 4)和Sox2(SRY-related high-mobility-group box gene 2)是胚胎早期发育和维持胚胎干细胞特征必须的转录因子，二者在消化道器官发育和肿瘤的发生、发展中起重要作用。Oct4主要表达于早期生殖细胞，随着生殖细胞的成熟其表达逐渐下调。Nichols等敲除小鼠的Oct4基因后，由于胚胎发育早期不能形成多能性的内细胞团，最终导致小鼠胚胎死亡。Sox2属于Sox家族，作为干细胞的另一个标志物，其主要功能是参与胚胎发育，维持中枢神经系统干细胞的活性。

本研究采用免疫组织化学SP法和流式细胞术检测Oct4和Sox2蛋白在食管鳞癌中的表达及相互关系，以期为深入研究食管癌发生、发展机制及寻找肿瘤新型靶向治疗方法提供依据。

1材料与方法

河北北方学院附属第一医院2012-01-01—2013-06-30切除存档的食管癌标本蜡块98例，患者年龄36~82岁，平均年龄(61.25±8.13)岁，中位年龄57岁；……
