程序性细胞死亡因子4与细胞色素C在上皮性卵巢癌中的表达及其关系

[摘要] 目的 研究上皮性卵巢癌组织中程序性细胞死亡因子4(PDCD4)和细胞色素C(Cyt C)的表达及其关系,探讨PDCD4诱导细胞凋亡的途径。 方法 收集2010年1月~2015年1月菏泽市立医院妇科62例手术切除的卵巢癌标本,采用免疫组织化学法检测上皮性卵巢癌石蜡标本中PDCD4和Cyt C表达情况,采用配对χ2检验分析PDCD4与Cyt C表达的关系。 结果 62例患者中33例(53.2%)患者PDCD4呈阴性表达,29例(46.8%)患者PDCD4呈阳性表达;40例(64.5%)患者Cyt C呈阴性表达,22例(35.5%)Cyt C呈阳性细胞表达。PDCD4蛋白表达呈阳性的患者Cyt C表达亦多呈阳性;Cyt C蛋白表达呈阴性的患者PDCD4表达亦多呈阴性,配对χ2检验结果显示,PDCD4和Cyt C的表达具有一定的相关性(χ2=5.652,P < 0.05)。 结论 上皮性卵巢癌中PDCD4和Cyt C表达密切相关,PDCD4可能通过线粒体途径引起Cyt C释放,启动凋亡程序,诱导细胞凋亡。

[Abstract] Objective To research expression of programmed cell programmed cell death 4 (PDCD4) and Cytc in epithelial ovarian cancer in epithelial ovarian cancer and its relationship, to investigate the pathways of PDCD4 induce apoptosis of pancreatic cancer cells. Methods 62 patients who received pancreatic resection from January 2010 to January 2015 in Heze Municiple Hospital were collected. The expression of Cyt C and PDCD4 in the samples was detected by immunohistochemistry, and the relationship between PDCD4 and Cyt C in pancreatic cancer was analyzed by applying paired χ2 test. Results Compared with the regular pancreatic tissues, the expression of Cyt C and PDCD4 in the cancer cells were significantly decreased. Among 62 samples, 37 cases (59.7%) had negative expression of PDCD4, 25 cases (40.3%) had positive expression of PDCD4; 45 cases (72.6%) had negative expression of Cyt C, 17 cases (27.4%) had positive expression of Cyt C. In general, patients who had positive expression of PDCD4 would have negative expression of Cyt C, and the patients who had negative expression of Cyt C would have negative expression of PDCD 4 as well. Paired χ2 test showed that, the expression of those two components were closely correlated (χ2=5.652, P < 0.05). Conclusion There is a close relationship between expression of PDCD 4 and Cyt C in pancreatic cancer. PDCD 4 may induce the apoptosis of pancreatic cancer cells through mitochondria.

卵巢癌的发病率居妇科恶性肿瘤的第三位,病死率居首位[1]。卵巢癌的特点主要是起病隐匿,早期缺乏典型症状和成熟的早期诊断方法,超过70%的患者确诊时已属晚期,约70%的患者在两年内复发[2],严重威胁广大妇女的生命。肿瘤的发生、发展需要细胞增殖能力增强和凋亡能力下降 。细胞色素C(Cytochrome C,Cyt C)从线粒体释放到细胞质是凋亡程序关键的一步,在凋亡机制中发挥着重大作用,是细胞凋亡过程中关键的细胞器之一,线粒体功能障碍导致Cyt C从线粒体膜间隙释放是多种细胞凋亡的普遍现象[3]。程序性细胞死亡因子4(programmed cell death 4,PDCD4)是新近发现的肿瘤抑制基因。通过抑制肿瘤细胞蛋白转录和翻译过程抑制肿瘤增殖。最近有文献报道,肺癌组织中的PDCD4表达明显下降或缺失,并与肺癌的恶性程度及不良预后有关[4]。……
