[摘要] 目的 检测Rab27A与Rab27B在3种人肝癌细胞系及1种永生化肝细胞系中的表达情况，初步研究Rab27A与Rab27B表达对肝癌细胞生物学特性的影响。 方法 采用RT-PCR及Western blot技术，检测Rab27A与Rab27B mRNA在3种人肝癌癌细胞HepG2、Huh-7、Li-7及1种永生化肝细胞系HL-7702中的表达。 结果 Rab27A与Rab27B在肝癌细胞系及原发性肝癌组织中存在差异性表达。Rab27A mRNA在3种人肝癌细胞系中表达明显强于1种人永生化肝细胞系；Rab27B mRNA在3种人肝癌及1种人永生化肝细胞系中均明显表达但无显著差异。 结论 Rab27A与Rab27B与原发性肝癌密切相关，可以为临床采取合理诊断措施提供有效的参考依据。

Expression of Rab27A and Rab27B in the four different hepatocellular carcinoma

[Abstract] Objective To investigate the expression of Rab27A and Rab27B in 3 human HCC lines and the immortalized hepatic cell line and its effect on biology characteristics of the cells. Methods The mRNA expression of Rab27A and Rab27B was detected by RT-PCR，and Western-blot in 3 human HCC lines of HepG2，Huh-7，Li-7 and the immortalized hepatic cell line of HL-7702. Results Rab27A and Rab27B were differentially expressed in cell lines and primary HCC tumors.The expression of Rab27A in 3 human HCC line was significantly better than that expressed in the immortalized hepatic cell line；the mRNA of Rab27B in 3 human HCC lines and the immortalized hepatic cell line were significantly expression but there was no significantly difference. Conclusion Rab27A and Rab27B expression are closely correlated HCC，it can provide the clinical reasonable diagnostic measures as valuable effective reference basis.

肝细胞癌（hepatocellular carcinoma，HCC）是消化系统最常见的恶性肿瘤之一，具有极高的侵袭性和转移性，出现典型症状时，往往已经为晚期。目前与HCC进展相关的因子包括癌胚抗原（CEA）、糖类抗原（CA199）、异常凝血酶原（DCP）等，同时Rab家族被证实与肿瘤密切相关。本文采用RT-PCR及Western blot技术检测了Rab27A与Rab27B在3种人肝癌细胞系及1种永生化肝细胞系中的表达情况，以期进一步研究其对肝癌细胞生物学特性的影响，为原发性肝癌的研究提供理论基础。

1 材料与方法

1.1 材料和仪器

3种人肝癌细胞HepG2、Huh-7、Li-7及1种永生化肝细胞系HL-7702均采购自中国科学院上海生科院细胞资源中心，并按照其建议的方法进行培养。20%胎牛血清、RPMI 1640培养基、DMEM高糖培养液购自GIBCO公司，DEPC购自Sigma公司，ECL显色试剂盒购自Thermo-fisher公司，NC膜购自Amersham公司，引物、Rab27A兔多抗、Rab27B羊多抗、HRP标记驴抗兔IgG、β-actin购自生工生物工程公司，Trizol、Taq PCR试剂盒购自TaKara公司，PCR反应仪采……
