[摘要] 目的 建立高效液相色谱法测定清热化毒丸中连翘苷含量的方法。 方法 采用Hypersil C18（4.6 mm×250 mm，5 μm）色谱柱；以乙腈-水（26∶74）为流动相，检测波长为277 nm，流速为1.0 ml/min。 结果 连翘苷进样量在0.2124～5.3100 μg （r=0.9994）线性范围内，线性关系良好，平均回收率为98.46%，RSD=0.86%（n=6）。 结论 该方法准确、重复性好，可用于清热化毒丸的质量控制。

The content of phillyrin of Qingre huadu wan determined by high performance liquid chromatography

[Abstract] Objective To establish the method of high performance liquid chromatography (HPLC) determining the content of phillyrin of Qingre huadu wan. Methods Hypersil C18（4.6 mm×250 mm,5 μm） chromatographic column was used.The mobile phase cinsisted of acetonitrile-water (26∶74),the detection wavelength was 277 nm and the flow rate of 1.0 ml/min. Results Phillyrin sample volume showed good linear relationship in the range of 0.2124-5.3100 μg (r=0.9994);the average recovery rate was 98.46%,RSD=0.86% (n=6). Conclusion The method is accurate with a good reproducibility,and suitable for quality control of Qingre huadu wan.

清热化毒丸是由连翘、青黛、黄芩、黄连、菊花、犀角粉等16味药炼制而成，收载于原卫生部药品标准·中药成方制剂第二册，具有清火化毒，消肿止痛的作用，主要用于小儿身热烦躁，咽喉肿痛，口舌生疮，皮肤疮疖，口臭，便秘等。原标准中只有薄层色谱法进行定性分析，而文献资料只有对黄连、黄芩之类的质量分析，没有针对连翘苷的定量分析，药理实验表明，连翘苷具有较强的抗病毒、强心、抗肝损伤性等活性，为了有效地控制药品的内在质量，笔者故建立对连翘苷的高效液相色谱测定法（HPLC）[1-4]。

1 仪器与试药

1.1 仪器

Waters e2695型高效液相色谱仪；Waters2489紫外检测器；Waters Empower色谱工作站。BP 211D电子分析天平（德国赛多利斯公司），9860 A型超声仪（费纳米科技发展有限公司）。

1.2 试药

连翘苷对照品（中国药品生物制品检定所，批号：110821-201112）；清热化毒丸（辽宁仙草堂药业股份有限公司，批号：130901，130301，130501）；乙腈为色谱纯，水为超纯水，其他试剂均为分析纯。

2 方法与结果

2.1 色谱条件

色谱柱 Hypersil C18（4.6 mm×250 mm；5μm）；流动相：乙腈-水（26∶74）；流速：1.0 ml/min；柱温：28℃；波长：277 nm；理论板数：按连翘苷计算≥3000[5-9]。

2.2 溶液制备

2.2.1 对照品溶液的制备精密称取连翘苷对照品适量，加甲醇制成每1 ml含0.2 mg的溶液，即得。……
