HPLC测定复方维生素注射液中维生素A、E、K的含量

【摘要】 目的 探讨高效液相色谱法(high performance liquid chromatography,HPLC)测定复方维生素注射液A、E、K1含量的可行性。方法 采用HPLC法,C18色谱柱(150 mm×46 mm,5 μm),流动相:甲醇-酒精(80∶20),检测波长270 nm。结果 维生素A棕榈酸酯在125~125.5 μg/ml之间线性良好(r=0.999 9),平均回收率为99.38%,RSD为1.36%;

维生素E在125~125.5 μg/ml之间线性良好(r=0.999 9),平均回收率为95.87%,RSD为0.95%。维生素K1在16.5~166.5 μg/m之间线性良好(r=0.999 8),平均回收率为98.08%,RSD为1.57%。结论 HPLC法测定复方维生素注射液维生素A、E、K1的含量专属性强,灵敏度高,准确性好。

【Abstract】 Objective To determine the contents in compound vitamins A、E、K1injection by high performance liquid chromatography(HPLC).Methods HPLC was used C18column(150 mm×46 mm,5 μm)was selected,the mobile phase was methanol-alcohol(80∶20).Detection was performed by UV with wavelength of 270 nm.Results A linearity of retinyl palmitate was obtained from 125 to 125.5 μg/ml with good correlation(r=0.999 9).The average recovery of retinyl palmitate was 99.38%,and RSD Was 1.36%.A linearity of vitamin E was obtained from 125 to 1251 μg/mlwith good correlation(r=0.999 8).The average recovery of vitamin E was 98.87%,and RSD was 0.95%.A linearity of vitamin K1was obtained from 16.5 to 166.5 μg/ml with good correlation(r=0.999).The average recovery of vitamin K1was 98.08%,and RSD was 1.57%.Conclusion The RP-HPLC is sensitive and accurate in determining thecontents in compound vitamin injection.

维生素(vitamin)是维持人体正常物质代谢和某些特殊生理功能不可缺少的低分子有机化合物,主要参与各种酶的组成,因其结构和理化性质不同,使其各具特殊的生理功能。复方维生素注射液可作为全营养静脉输液的添加剂,以满足每日的维生素需要[1]。近来有系统测定维生素的报道,但多数是报道纯品维生素的分离分析。本文应用高效液相色谱法(high performance liquid chromatography,HPLC)测定复方维生素注射液A、E、K1含量,现作报告。

1 仪器与试剂

仪器:岛津LC-10A高效液相色谱仪系统,包括LC-10ATvp泵,SPD-10Avp紫外检测器,C-R8A色谱数据处理仪。色谱柱:以十八烷基硅烷键合硅胶为填充剂(150 mm×46 mm,5 μm),不锈钢柱(Shin-pack CLC-ODS)。试剂:甲醇、酒精均为分析纯。维生素A棕榈酸酯对照品[由巴斯夫(中国)有限公司提供];维生素E对照品、维生素K1对照品(由中国药品生物制品检定所提供)。复方维生素注射液样品批号:041204,041206,041208

(由上海第一生化药业有限公司提供)。

2 方法与结果

2.1 色谱条件与系统适应性试验 色谱柱:shin-pack CLC-ODS C18柱(150 mm×46 mm,5 μm)。流动相:甲醇-酒精(80∶20),检测波长270 nm,流速0.5 ml/min。进样量20 μl。在上述条件下,维生素A棕榈酸酯、维生素E和维生素K1的保留时间分别约为39.98、23.96、18.41 min。……
